| ObjectivesThis research through three ordered experiments to approach the effect of the Chinese medicine " Tonic Liver Mixture " to acute liver failure rats which have Hepatocyte transplantation.Experiment 1: In order to find out the animal model closer to the human virus hepatitis induced acute liver failure,and use chemical and biological detection and pathological examination to confirm the reliability and stability of the model.Experiment 2:In order to explore the effect of Chinese medicine "Tonic Liver Mixture " on the liver cells of rat in vitro cultured.Experiment 3:In order to explore new methods of treatting acute liver failure,we use the treatment of hepatocyte transplantation unit Chinese medicine " Tonic Liver Mixture " on base of the acute liver failure model in rats.MethodsExperiment 1:We have adopted TAA and lipopolysaccharide(LPS) induced by different combinations of acute liver failure in SD rats.Experiment 2:Using the technique of liver cell culture and blood serum pharmacology,to compare the liver cells growth and proliferation in three different culture media-foundament culture medium,Chinese Angelica+ foundament culture medium," Tonic Liver Mixture "+ foundament culture medium,such as the role of the situation.Experiment 3:Dividaturing 80 I-line rats which handled by the method of experiment 1 into three arrays- transplantation group,transplantation+ Chinese medicine group,control group,to compare their persistence and biochemical indicators.ResultsExperiment 1:results show that all of merely TAA twice and LPS second instead can establish the animal model of acute liver failure.Among of total,the way of injecting with 3%TAA 300mg/kg in the first day and LPS 5μg/kg in the next day established a stable animal model of acute liver failure,it is survival window phase is more than 48h,it is easy to develop the therapeutic research.It is a relatively ideal animal model.Experiment 2:The results show that traditional Chinese medicine " Tonic Liver Mixture " promoted liver cells to grow and proliferate in vitro,and significant prolonged the survival time in vitro,suggesting that " Tonic Liver Mixture " may promoted liver cells to produce a marked effect and form the dynatron effect of nourishing and protecting liver.Experiment 3:The results show that although hepatocyte transplantation did not significantly reduce liver cell injury,but can significantly compensate the metabolic function of liver cells,reduce the concentration of serum direct bilirubin and blood ammonia,improve the level of serum albumin,shortenedprothrombin time,significantly improve the survival rate of five days. Combination of Chinese medicine " Tonic Liver Mixture ",improve the fifyh day's survival rate furthermroe,reduce the concentration of direct bilirubin and ammonia,improve the level of serum albumin, shortened prothrombin time more significantly,and also a significant reduction for the level of ALT and AST in serum, suggesting that the Chinese medicine " Tonic Liver Mixture " hepatic cell transplantation for acute liver failure to save the lives of affected persons have significant therapeutic value.ConclusionsUs through the above three experiments,to build a relatively ideal model of acute liver failure in rats,to verify the " Tonic Liver Mixture " promoted the growth of liver cells in vitro based on cytoecology,initial confirmed the optimize and supporting role of Chinese medicine " Tonic Liver Mixture " when hepatocyte transplantation in the treatment of acute liver failure,this subject lay the foundation for clinical trials.
