Endometrial cancer is also called as corpus carcinoma,which is a series of malignant epithelium tumors that primarily stem from endometrium.Among the total, most of it is adenocarcinoma endometria that stems from endometrial glandular organ. Endometrial cancer is one of three common malignant tumors in the female genital tract,counts 7%or so in the female carcinomas,and 20%~30%in malignant tumors in female genital tract.Endometrial cancer is frequent in the old women.The incidence of endometrial cancer is highly increasing in the young women in recent years.But during recent 20 years because duration of life becomes longer and longer,eider women become more and more,that is said the number of patients at the high-incidence age of endometrial cancer is increased,adding up to using exogenous estrogen extensively, especially without progestogen-resistance.Without progestogen-resistance endometrial hyperplasia will happen and atypical hyperplasia will result in cancerization,thus in the world the morbidity of endometrial cancer shows ascending tendency.During the period of the recent 10~20 years,the morbidity of endometrial cancer has maintained twice of the one of the middle of 70's.It threatens women's lives seriously.Studying its mechanism in tumorigenesis and seeking for effective method in therapy have become important problems for study.Hpa is endo-B-glucuronidases,it plays an important role in tumor's growth,invasion,metastasis and angiogenesis,and MVD is a reliable indication to evaluate angiogenesis of tumors.The research of MVD and expression, clinical significance of Hpa in Endometrial cancer is not reported.Materials and methodsWe selected 33 of endometrial carcinomal(FIGO,stageⅠ:18cases,stageⅡ:10 cases,stageⅢ-Ⅳ:5cases) who were enrolled from 1992 to 2002 year in the Shengjing Hospital of China Medical University.They were not treated before operation and all the patients had full search for data,the age is from 32 to 70 years old,the average age is 50.Twelve cases normal endometrium and nine endometrial atypical hyperplasias were also studied as control group.All specimens were stained immunohistochemically for cd34 anti Hpa identified immunohistochemically with specific antibody.The method observed slices under 400 double light microscope,photoed under digital camera,input 5 fields random selected into MetaMorph,converted staining intensity of cells to quantification index,calculated opital density.MVD was indicated by mean number of vessels of three high power fields.All experimental data were analyzed with ANOVA and processed by SPSS11.5 There was a statistical significance when P<0.05. The relationship of the two variables is analyzed by Pearson correlation analysis method.ResultsThere were significantly differences of Hpa in normal endometrium,atypical hyperplasia of endometrium and endometrial cancer(P<0.01).It's highest in the endometrial cancer and lowest in the normal endometrium.Hpa expression was significantly correlated with two clinic pathological features:histological type(P<0.01), clinic stage(P<0.01).Hpa expression has nothing with the degree of differentiation(P) 0.05).There were significantly differences of MVD in normal endometrium,atypical hyperplasia of endometrium and endometrial cancer(P<0.01).It is highest in endometrial cancer and lowest in normal endometrium.There were significantly differences of MVD in grade 1 and grade 3.It is highest in grade 3 and lowest in grade 1.MVD was significantly higher in stageⅢthan in stageⅡ.MVD in normal endometrium,atypical hyperplasia of endometrium and endometrial cancer was positively related Hpa in normal endometrium,atypical hyperplasia of endometrium and endometrial cancer.There was a significant difference between two groups(Pearson correlation coefficient = 0.8,P<0.01).Conclusions The expression level of Hpa in EC is significantly higher than in normal endometrium.and the expression level is increasing with the development of pathologic lesion,which indicated they may have important function with the pathogenesis and plasty of EC.MVD was significantly higher in EC than in normal endometrium and with the development of EC MVD increased which indicated angiogenesis may play an important role in development of EC.MVD may be an valuable indicator in prognosis judgment of EC.MVD may be an valuable indicator in prognosis judgment in patients with EC.Meanwhile,MVD in tumors with positive expressions of Hpa was significantly higher than that with negative expression,which indicated that they may promote the angiogenesis of EC.
