Recent years, a medicinal pteridophyte plant Huperzia serrata (Thunb.)Trev. has become a research hotspot due to its active ingredient Huperzine A. Huperzine A has good effects on cholinesterase activity inhibition, and Alzheimer's disease treatment. In this study, endophytic fungus were isolated from Fujian Huperzia serrata. The huperzine A content in Huperzia serrata (Thunb.)Trev. and endophytic fungus was deteceted by HPLC. The results were as follows:1. The HPLC system for huperzine A: Waters C18 column, 150mm×4.6mm, 5μm; methanol- 0.08mol·L-1 ammonium acetate buffer (30:70, pH 6.0 ); 1 ml·min-1; 310nm; 30℃. The content of Hup A in Huperzia serrata stems was 197.80μg/g, and 87.54μg/g in leaves.2. The fungus haustorium was found in the stems as well as leaves in Huperzia serrata. It indicated that the endophytic fungi was existed in Fujian Huperzia serrata.3. 5 strains of endophytic fungus were isolated and identified from the stem and leave of Huperzia serrata. And were identified as Colletotrichum sp., Rhizoctonia sp., Fusarium sp., Glomus sp., Alternaria sp.. It covered 2 subdivsions, 3 classes, 5 orders, 5 families, and 5 genus.The quantity of endophytic fungus in leaves was more than that in stems in Fujian Huperzia serrata.4. The optimum culture temperature of these 5 endophytic fungus were 25℃; and grew better under light condition.5. Small amount of Hup A was found in the bacterial liquid of Colletotrichum sp. by HPLC.
