A medicinal plant Huperzia serrata has been selected to isolate its endophytic fungus and its secondary metabolites were observed. Endophytic Fungi was isolated from healthy Huperzia serrata with conventional isolation method. Results indicated that ninty-four Endophytic fungal strains from different parts of Huperzia serrata varied, and among which 76 from the leaves,12 strains from the stems and the rest from roots, there is not one strain isolated from sori. Antagonistic activities to some plant pathogens were surveyed and results showed that nine strains have inhibited effect on Sphaerotheca fuliginea(sf).There is a strain of Endophytic fungi in Huperzia serrata have the more than 80% effect on sf and 2 strains over 75%.The results of HPLC detection analysis indicated that probably a strain has produced HupA and is mostly close to standard sample Hup A.We choose A35b strain as researching the optimal culture conditions. The conditions for the growth of A35b strain were as follows: the optimal temperature was 27℃; the optimal pH was 5.5;the optimal illumination was alternate between day and dark, he optimal nitrogen and carbon source were east ointment and maltose.
