ObjectiveTo investigate the effect of Qi gui yi mai ling(QGYML) on protein kinase C during acute myocardial ischemia reperfusion in rat myocardial cell,then study the mechanism of the protective effect of Qi gui yi mai ling on myocardium during ischemia reperfusion injury.MethodsThe experiment divide all of the 56 wister rats into 7 groups at random: normal saline group,sham operation group,ischemia reperfusion group, ischemic preconditioning group,QGYML high dose group(2g/kg),QGYML low dose group(lg/kg),and Isosorbide Dinitrate group.The ligation/ recanalization ischemia reperfusion injury were induced by ligation of the left anterior descending branch of the coronary artery,take cardiac muscle under ligature for reservation.The expression of protein kinase C in myocardial cell of the 56 rats was detected by immunohistochemistry.ResultsAfter ischemia reperfusion injury,all groups have show different activity expression of PKC.Compared with ischemia preconditioning group,the PKC activity remained remarkable difference change in QGYML high dose group(p<0.05);there is no significant difference in statistics between QGYML high dose group and ischemia preconditioning group (p>0.05).Then the expression of PKC was significant difference in QGYML high dose group and QGYML low dose group(p<0.05),Among the total,there is no significant difference in statistics between QGYML high dose group and lsosorbide Dinitrate group(p>0.05).Conclusion1.Qi gui yi mai ling can protect ischemia reperfusion injury from elevating the activation of PKC in cell membrane.2.Qi gui yi mai ling can elevate anti-myocardial ischemia,then relieve ischemiareperfusion injury,and has the same effect as Isosorbide Dinitrate, so it has the effect of protecting cardial muscle as pharmacological preconditioning.
