ObjectiveThe topic intends to observe whether preconditioning oftraditional Chinese medicine compound Ruan mai ling can inhibit theinflammaion during myocardial ischemia reperfusion injury withmodels of rats on the basis of previous studies.MethodsBefore surgery,the rats are connected limb lead II to conduct ECGmonitoring,and ECG abnormalities are excluded.The enrolled rats arenumbered from1to48and divided into six groups using a randomnumber table. The six groups are as follows:sham operation group(sham group), model group (I/R group), Ruan mai ling low-dose group(RMLL group), Ruan mai ling moderate-dose group (RMLM group),Ruan mai ling high-dose group (RMLH group) and dexamethasonegroup(DSMS group).There are eight rats in each group.The coronaryleft anterior descending branches of treatment groups and modelgroup are ligated30mins,and then the ligatures are loosened2h toestablish myocardial ischemia-reperfusion model.The coronary leftanterior descending branches of sham group are only stringed but notligated.After HE staining,observe the pathological changes ofmyocardial tissue under light microscope of400times;detect the protein expression levels of NF-κB p65in myocardium nucleus byimmunohistochemical;detect the expression levels of TNF-αinmyocardial tissue by Western blotting;detect the levels of IL-1β inserum by enzymelinked immunosorbent assay;detect the activity ofMPO in myocardial tissue by colorimetry.Results1、 Changes of TNF-α、 MPO、 NF-κB p65、 IL-1β:Compared with theshamgroup,TNF-α expression,IL-1β concentration,NF-κB expressionand MPO activity of the rest five groups increase(▲P<0.01or▲P<0.05);Compared with the model group,TNF-α expression,IL-1β concentrat-ion,NF-κB expression and MPO activity of RML low, Moderate andhigh dose group and dexamethasone group decrease (△P<0.01or△P<0.05);The difference among RML groups are obvious (P<0.05or*P<0.05);There is no significant difference between the RML high dosegroup and the dexamethasone group(*P>0.05).2、 Pathological changes in myocardial tissue of rats: under the lightmicroscope,we can see myocardial fibers arranged in neat rows,clearstripes,no necrosis and swelling fracture,no neutrophil infiltration inthe sham group;myocardial fibers swelling,some stripes disappeared,partial bleeding,myocardial cell necrosis, infiltration of neutrophils inthe model group; slight swelling and still orderly arrangement ofmyocardial fibers, a small amount of neutrophil infiltration in thedexamethasone group;only a few of myocardial cells swelling,minimalinflammatory cell infiltration in the dexamethasone group.ConclusionsPreconditioning of Ruan mai ling can inhibit the inflammation inrat myocardial ischemia-reperfusion injury,reduce the injure caused by reperfusion,and play a protective role with myocardial tissue.And thiseffect is dose-dependent.
