The Effect Of Neizhang Pill On T Lymphocyte Subsets Following Anti-Sperm Antibody-Positive In Adult Mice

Objective:This study using the method of active immunization to establish AsAb positive animal models,to observe the effect of neizhang pill on T lymphocyte subsets(CD3+,CD4+,CDS+) biological activity in mice peripheral blood and decidual,and further explore the pathogenesis of autoimmune abortion and the mechanism of neizhang pill on cure autoimmune abortion.Method:Refer to the method of Anne Lai to AsAb positive animal model.Female mice,the total number of 60.All were randomly divided into 4 groups,every group have 10.Chinese medicine group(neizhang pill 19.44g·kg^-1·d¹);Western medicine group(prednisone acetate tablets 1.35mg·kg^-1·d^-1);model group and blank group(such as dose of distilled water).The week after the first immuni-zation beginning filled stomach until 3 week.Executed after 9 days of pregnancy, and observed the outcome of pregnancy;Measured by ELISA in peripheral blood of anti-sperm antibodies;by flow cytometry analysis of mouse peripheral blood and decidual T lymphocyte subsets in the expression.Results:1.The antisperm antibody animal model was established successfully.2.Neizhang pill can effectively inhibit the emergence of anti-sperm antibodies.3.Anti-sperm antibodies in mice peripheral blood and decidual,CD4+levels and CD4+/CDS+ ratio increased significantly.Prompted AsAb positive mice exist systemic and uterine immune disorder.4.Neizhang pill can reduce abnormal increased CD4+,CD4+/CDS+ content of the anti-sperm antibody-positive in mice peripheral blood and decidua. Thus the introduction of Neizhang pill may be impaired in T-lymphocyte subsets to correct the imbalance ratio,the restoration of CD8+ and CD4+ T lymphocyte immune response in the body in the process of coordinating role in or- der to regulate systemic and local immune disorder state of the uterus.5.Neizhang pill can be markedly improved the number of mice born,and more effective than prednisolone acetate.
