OBJECTIVE: By the research on the extraction, purification and column chromatography process of flavonoids in Scutellaria baicalensis, we select the best methods for the chemical standard sample preparation of flavonoids in Scutellaria baicalensis. At the same time,by the study on the different harvesting period, different parts and different prepared medical materials of the Scutellaria baicalensis in Shandong province for the quality control with the techniques of chemically qualitative and quantitative analysis,the purpose of this thesis is to provide scientific data to establish the quality control method and evaluation standard of Scutellaria baicalensis.METHODS: By the Single-factor study, orthogonal design and the use of HPLC quantitative analysis,we determine the best process of flavonoids' extraction and the baicalin's purification in Scutellaria baicalensis . Five active components are isolated from Scutellaria baicalensis by the use of column chromatography technology for separation and purification. It is performed for the further research on the flavonoids of the different harvesting period, different parts and different prepared medical materials in the Scutellaria baicalensis in Shandong province by HPLC and modern key technologies of Chinese medicine quality control.RESULTS: 1.The established extraction process of the flavonoids in Scutellaria baicalensis is efficient, feasible and reproducible . And the extraction rates of baicalin, baicalein, wogonin can reach 90%,75%,80% separately. 2. The transfer rate and purity of the established baicalin's purification process increase significantly than conventional technology. The transfer rate and the purity of Baicalin can reach 70% and 85% by the purification process. 3.The column chromatography technology for separation and purification of the flavonoids in Scutellaria baicalensis is simple, reasonable and feasible,by which we can get the four active ingredients and the purity of them higher than 98% in turn. 4. In different stages of harvesting Radix Scutellaria, baicalin, wogonoside, wild baicalin, wogonin content and total flavonoids are the highest and in September 17 harvest samples. The content of baicalein and oroxylin-A is decreases with delayed harvest period 5. The content of baicalin in root bark is higher than that in root heart.The content of baicalin in part of the ground (stems, leaves) is far lower than that in roots, but content of scutellarin is up 10 times than baicalin. 6. The content of baicalin after drying, steam or boil is higher than that after natural dry, steamed slices lightly higher than boiled slice, the content of baicalin decreases and the content of baicalein and wogonin increase significantly.
