Scutellar baicalensis Georgi is known as one of the precious ancient Chinese herbal medicine.There are various of bioactive compounds in Scutellar baicalensis Georgi.And it has been proven by a large number of pharmacological and clinical studies that Scutellar baicalensis Georgi has several physiological and medicinal functions such as anti-cancer activity,control blood sugar,immune regulation,anti-bacterial activity,and anti-inflammatory.Furthermore,recent investments home and abroad show the other physiological functions of Scutellar baicalensise Georgi,which presents great potential for application.Gastrodia,is another famous ancient Chinese medicine,with good various of physiological and medicinal functions such as antioxidant activity,analgesic,improved learning and enhance memory,enhance nonspecific immunity and cells immune.Especially the high antibacterial activity of Gastrodia elata,which has been favored in recent yearsIn this study,in order to comprehensively considerate of the anti-bacterial activity of Scutellaria baicalensis Georgi,single factor experiments were used to optimize the extraction temperature,extraction time,liquid material ratio of Scutellaria baicalensis Georgi.Followed by Box-Behnken design experiment scheme,response surface analysis method to extract the extraction temperature,extraction time,liquid material ratio of the optimal value.Based on single-factor optimization strategy and Box-Behnken design experiment,the optimal extraction of Scutellaria baicalensis Georgi was obtained.The extraction temperature was 61 ?,extraction time 2 h,liquid material ratio 13: 1(mL: g).Under the conditions of this process,the effective antimicrobial composition of Scutellaria baicalensis Georgi,the best antibacterial value was 13.062 mm,which was 10.53% higher than that before optimization.Mutation technique was used to optimize the extraction temperature,extraction time,salt concentration of Gastrodia protein.Based on single factor optimization and Box-Behnken design experiment,the optimal extraction conditions were as follows: extraction temperature 77 ?,extraction time 4 h,salt concentration of 0.8%.Under the conditions of this process,the best antibacterial value of Gastrodia protein was 89.08%,which was16.30% higher than that before optimization.In addition,the ext-raction of Gastrodia protein had good antibacterial activity to Candida albicans,Esc-herichia coli,Pseudomonas aeruginosa,Staphylococcus aureus.The antibacterial ra-tes were 20.89%,22.02%,22.71% and 16.73% respectively.Furthermore,the stabil-ity test of the antibacterial activity of prontein of Gastrodia was test,and the result showed that the antibacterial activity of Gastrodia protein was stable.Also the safety of Gastrodia protein concentrate assessment was study,the result showed that Gastrodia protein concentrate was safe,and without any side effects.In order to obtained a natural compound antibacterial spray,the extraction liquid of Scutellaria baicalensis Georgi and Gastrodia protein concentrate was as the main ingredient,the Dendrobium skin,purple flowers and other traditional Chinese medicine extraction liquid were added into the natural compound antibacterial spray.In addition,the antibacterial activity of spray was tested,and the results showed that the natural compound spray has good antibacterial activity.Furthermore,skin irritation and pathology were used to tested the safety of spray,the results found that natural compound spray was safety,and without irritation to cavia porcellus skin.
