Glucocorticoid amplifies IL-2-induced expand the ratio of CD4+CD25+to CD4+CD25-and inhibit the allogeneic reactionObjective:1.To determinate the effect of the short-term administration of DXM and IL-2 in vivo on the mixed lymphocyte reaction.2.To establish the method to expand the ratio of CD4+CD25+to CD4+CD25- by DXM and IL-2 in vitro.Methods:1.Seven days after administration of DXM(5mg/kg/day,3days) and IL-2 (400,000IU/mouse/day,3days) to C57BL/6N mice,mixed lymphocyte reaction was taken by fresh spleen cells from C57BL/6N mice and untreated BALB/c mice.2.In vitro,fresh spleen cells from untreated C57BL/6N mice were cultured with IL-2,and added DXM 4 hours later.The cells were made Flow cytometry analysis 20 hours after coculture.Results:1.In vivo,short-term simultaneous administration of DXM and IL-2 markedly expanded functional suppressive CD4+CD25+regulatory T cells in murine spleen and inhibited mixed lymphocyte reaction obviously.2.In vitro,the combination of DXM and IL-2 can selectively enhance the proportion of CD4+CD25 regulatory T cells,the ratio of CD4+CD25+to CD4+CD25-was also changed.Conclusions:The administration of DXM and IL-2 inhibit mixed lymphocyte reaction.And this because the combined use of DXM and IL-2 expand the ration of CD4+CD25+to CD4+CD25-.
