Objective:To investigate the expression of interleukin-3 receptor alpha(CD123) on bone marrow cells in acute myelocytic leukemia(AML) and its clinical significances.Metheds:By means of Fluorescence-activated cell sorer(FACS) and semi-quantity reverse transcripition polymerase chain reaction(RT-PCR),the expression of IL-3Rα(CD123~+) protein on CD34~+CD38~- cells and mRNA in BMMNCs of 71 AML patients of Tianjin Medical University General Hospital from March 2008 to february 2009 and 16 normal controls were detected respectively,Then the correlation between IL-3Rαand the clinical stages of AML were analyzed.Results:1.CD34~+CD38~-CD123~+/CD34~+CD38~- and IL-3RαmRNA in BMMNC of 38 deno-vo or relapsed AML patients were(0.5602±0.2834) and(0.7819±0.4420) respectively.They were higher than those of control group[(0.0296±0.0491),(0.0722±0.1754 )](p<0.05).2.CD34~+CD38~-CD123~+/CD34~+CD38~- and IL-3RαmRNA in BMMNC of 33 AML patients in remission were(0.3711±0.2880) and(0.3144±0.4970) respectively.They were significantly different from those of control group(p<0.05) and also lower than those of de no-vo and relapsed AML patients(P<0.05).3.CD34~+CD38~-CD123~+/CD34~+CD38~- and IL-3RαmRNA in BMMNC of M2,M3,M4,M5 subtype patients from 38 deno-vo or relapsed AML were[(0.5593±0.3175),(0.8816±0.2448)],[(0.3975±0.3599),[(0.3908±0.5527)],[(0.5369±0.2780),(0.7882±0.4259)],[(0.5858±0.2611),(0.7518±0.5435)] respectively,they have no significantly difference(P>0.05).4.14 AML patients were followed up.and 7 of them were from deno-vo to remission,their CD34~+CD38~-CD123~+/CD34~+CD38~- and IL-3RαmRNA decreased from[(0.6321±0.3548),(0.7084±0.5218)]to[(0.4101±0.2929), (0.2431±0.4153)](p<0.05);3 of them were from remission to relapsed,their increased from[(0.6253±0.3545),(0.3150±0.1462)]to[(0.8503±0.8809),(1.056 ±0.6542)](p<0.05);and 4 of them were in remission constantly,their CD34~+CD38~-CD123~+/CD34~+CD38~- was(0.1949±0.1562),and their IL-3RαmRNA could not be detected.Conclusion:①The expression of IL-3Rαon BMMNC of AML patients increased at on setting or relapsing,it has no correlation with AML subtypes;②The expression of IL-3Rαof AML patients increased at on setting or relapsing,and decreased in remission,IL-3Rαgene might play an important role in AML malignant clone formation;Clinically It might be taken as a new parameter for the evaluation of the chemotherapy,response prognosis and relapse of AML.
