Objectives:The aim of this study was to detect the expression levels of IL-17 and IL-23 in Bone marrow of acute myeloid leukemia patients and understand the effect of IL-I7/IL-23 axis on immune regulatory mechanisms and role of the disease.Methods:Collected 39 AML patients who were diagnosed and treated in the department of Hematology of the first affiliated hospital of Kunming Medical University from October 2015 to December 2016 as a patient group,15 patients suffered anemia or other benign and non-immune diseases of blood system as control group.The fresh bone marrow was collected in the both AML group and control group.The IL-17mRNA and IL-23mRNA in bone marrowmononuclear cells of AML group and control group were detected by Real-time quantitative reverse transcription-polymerase chain reaction.At the same time,we collected the clinical data of the patients with AML.All test data was analyzed by the SPSS 17.0 statistical software.P<0.05 was statistically significant.Results:1.The expression level of IL-17mRNA in AML group before therapy and after treatment was lower than that in controls(P=0.001,P<0.05)(P=0.012,P<0.05).The patients in AML group before therapy was lower than the group of post treatment(P=0.0.07,P<0.05).The expression level of IL-17mRNA in patients with AML who achieved remission was higher than that in patients with AML before therapy(P=0.022,P<0.05).2.The expression level of IL-23mRNA in AML group before therapy and after treatment was lower than that in controls(P=0.000,P<0.05),(P=0.005,P<0.05).The patients in AML group before therapy was lower than the group of post treatment(P=0.011,P<0.05).The expression level of IL-23mRNA in patients with AML who achieved remission was higher than that in patients with AML before therapy(P=0.047,P<0.05).3.The expression level of IL-17mRNA and IL-23mRNA in the patients with AML was positively correlated(r= 0.815,P=0.000,P<0.05).The expression level of IL-17mRNA and IL-23mRNA in the patients with AML showed little correlation with gender?age?the number of WBC before therapy?ECOG score and LDH(P>0.05).Conclusions:IL-17 and IL-23 may be involved in the pathogenesis and development of AML.They may be helpful to judge the severity of the disease,efficacy and prognosis.And they may influence each other in this process through synergistic effect.
