The Effect Of Guiling Xiaoyi Decoction Drug Serum On The Morphology And The Secreting Of IL-6 IL-8 Of Eutopic Stromal-cell In Endometriosis

Endometriosis is a common intricate gynecological disease defined as endometriotic tissues growing outside the normal uterine cavity. This benign disease has confused clinicians for a long time, which behaves like malignant disease. This paper puts emphasis on the study of the theoretical foundation about the treatment by Guiling xiaoyi Decoction. We initially establish the in-vitro cultural method of purified stromal-cell, which provides an experimental model for the survey of the effect of Guiling xiaoyi Decoction on endometriosis and its mechanism of action.1,The study of the theoretical basis of the treatment of Guiling xiaoyi decoction for dometriosisThere is no term corresponding to endometriosis in traditional Chinese medical . The cardinal symptoms of endometriosis correlate with those of algomenorrhea, abdominal mass and infertility in traditional Chinese medicine. The disease is due to malfunction of the dominating reproduction of the kidney, overacting in sexual intercourse during menstruation and postpartum, and then leading to injury the Chong and Ren meridians and failure of the functions of storing and discharging of the uterus. The kidney's function being not sufficient is the radical factor of occurrence of the disease. Blood stasis is the pivot of occurrence and development of the disease. So the treatment for endometriosis should be based upon the concept of reinforcing warming kidney and dispersing blood stasis. Guizhi fuling pill originates from jinkui yaolue. Zhongjing Zhang ,the author of this book,designed this prescription to mildly remove mass in the abdomen,disperse blood stasis in order to prevent the abortion which was due to the abdominal mass endangering the fetus.If the prescription was utilized mainly to remove blood stasis and disperse the mass in the abdomen,there might be some shortage of the strength of the drugs in the prescription. On one hand, more drugs having the property of promoting blood circulation and resolving blood stasis should be added to the prescription. On the other hand, the prescription should be modified in accordance with syndrome differentiation. So Guiling xiaoyi Decoction was designed which combines the Guizhi fuling pill with the herbs Xianlingpi to strengthen warming kidney, and Xuejie to help dispersing blood stasis. The above mentioned notion forms the theoretical foundation of our vitro experimental study of the treatment of Chinese materia medica of warming kidney and dispersing blood stasis for endometriosis.2,In-vitro culture method of stromal-cell established initiallyWe collected endometria from patients who suffered from endometriosis and recived operation from April in 2007 to October in 2008. Stromal-cells were isolated from it and went down to the future subculture,the proliferative condition were observed by microscope,and purity coefficient of cell was characterized with immunohisto- chemistry method.As a result, the proliferative conditions of stromal-cells cultured in-vitro mainly with the method of grit separation were good. During observation, cells were all simple interstitial ones, went down to 4-5 generations and could survive 25-30 days. It can be show from the result of immunohistochemistry that the purity coefficient achieve percent 95 above.It can be seen from the result that under our experiment condition the stromal- cell cultured in-vitro has satisfactory ability of vegetation. The stromal-cells isolated from both endometriosis and non-endometriosis can be successfully cultered ( P>0.05). The menstrual cycle has an influence on the stromal-cell's survival rate and the ability of growth (P<0.05).An effective in-vitro cultured cell model can be provided to reveal pathogenesy of endometriosis and study the pharmacodynamics.3,The effect of Guiling xiaoyi decoction on appearance of stromal-cells and IL-6,IL-8 from cell culture supernatant fluidWe made serums of different dosage drugs of Guiling xiaoyi Decoction, interfered the stromal-cells cultured in-vitro which were extracted from the parents suffering endometriosis with the serums, then observe their morphologic change by the inverted microscope, and collected supernatant liquid of cell culture at the time of 24h,48h,72h,respectively detected the concentration of IL-6,IL-8 by the method of ELISA.From morphology observation, we can initially confirm that the serums of Guiling xiaoyi Decoction can efficaciously suppress the growth of stromal-cell cultured in-vitro,and this suppress depends on the concentration of the drug serums. Besides, Guiling xiaoyi Decoction can cut down externalization of IL-6 and IL-8 from the cells cultured in-vitro obviously. The observations in this paper strongly suggest that adjusting of immunization, restraining inflammatory topo-reaction of endometrial, abating adherence at pelvic cavity, reducing the invasion and proliferation capacity of ectopic cells and inhibiting the growth of neovascularization might be the effects of Guiling xiaoyi Decoction to control mobility and development of endometriosis.
