Objective:To study the expression of GnRHⅡin endometriosis and the effect on cells apoptosis on culture endometrial stromal cells in vitro.Methods:Thirty ectopic and thirty eutopic biopsy endometrial specimens were obtained from women with untreated endomentriosis and 40 controls were studied,which came from August 2007 to April 2008 in the Second Xiangya Hospital of Central South University.The expression of GnRHⅡprotein were tested by immuneohistochemistry, the endometrial stromal cells were isolated,cultured and identified from ectopic endometrium and eutopic endometrium tissues.Then,to detect the cell apoptosis rato by Hoechst staining and flow cytometry after endometrial stromal cells In vitro were used different concentration (0,10-10 M,10-8 M,10-6 M) GnRHⅡ.Results:(1) The expression of GnRHⅡprotein positive immunization staining is located in endometrial stromal cells and glands cytoplasm,(2) The expression of GnRHⅡprotein was lower in ectopic endometrium than eutopic endometrium of women with endometriosis(P<0.05),and were lower in the eutopic and ectopic endometrium with endometriosis than the nomal endometrium of women without endometriosis(P<0.05).(3) The expression of GnRHⅡprotein was lower in endometrial hyperplasia than that of in secretory phase in each group,but were significant difference only in nomal endometrium not in eutopic and ectopic endometrium.(4) In vitro culture, the apoptosis cell of endometrial stromal cells can be observed after GnRHⅡtreatment(0,10-10 M,10-8 M,10-6 M),The Hoechst dye showed that ectopic and eutopic endometrial stromal cells can be induced apoptosis:nuclear condensation into shape.The rate of apoptosis(%) is (2.58±0.73,4.90±0.41),(8.72±1.59,12.30±2.13),(19.90±2.37, 29.40±4.05),(36.10±2.36,52.30±5.42)(P<0.05),The flow cytometry test results showed that ectopic and eutopic endometrial stromal cells can be induced apoptosis.The rate of apoptosis(%) is(2.31±0.26,5.40±0.39,(10.20±1.29,14.50±2.17),(25.70±3.22,36.20±4.64), (59.70±6.63,72.30±8.12)(P<0.05),the apoptosis rate of ectopic endometrial stromal cells was higher than eutopic endometrial stromal cells(P<0.05),and were dose-dependent.Conclusion:(1) The lower espress of GnRHⅡmay cause endometriosis.(2) The expression of GnRHⅡwas related to the menstrual cycle in normal endometrium,there was not difference in EMs.(3) GnRHⅡenhanced stromal cells apoptosis in dose-dependent in vitro for the patients with EMs,it suggested:Exogenous GnRHⅡmay play an important role in treatment of the patients with EMs.
