The supply of calculus bovis has been unable to meet the demand, because of the low yield of natural ox gallstone. In the 1970s, many domestic researchers had been dedicated on cultivating calculus bovis in vitro and in vivo and successed. But the output and quantity of natural ox gallstone is not stable enough, for the reason that the mechanism how the gallstone formed is still not clearly understood, and it is quite hard to take completely control of the output and quality of ox gallstone, eg. the effect of the protein in bile in forming gallstone and dynamically observed the change of the key component of bile and pattern of gallstone formed during the process of formation is studied, through the trial of formation of bezoar in the external calculus bovis generators and in gallbladders of goat mainly based on electrophoretic technique,chromotography techniques. Six trials are in this experiment, as following:Ⅰ: Acclimatization of bile-resistant E.coli and electrophoretic analysis of protein in goat and cattle bileE.coli was inoculated in bouillon culture-medium with cattle-bile gradually augmented to make it be able to grow in bile; the bacterium can survival and multiply in bile, and the content is 108CFU/ml. Though the electrophoretic analysis of protein in goat and cattle bile with Markers in the same gelatin by Department of gel image analysis instrument and analysis system, It was found that they contain the same straps in the tracks; but the concentration of proteins with the same molecules were different.Ⅱ:Irradiation conditions of bile irradiation sterilization and the change in physical and chemical properties of bile before and after irradiationIrradiating bile using by Co60, sterilization rate was 98% when exposure dose was 5KGy, and the sterilization rate was 100% when exposure dose was 15KGy. The number of the straps see no changes and here are no significant changes of the optical density accumulation.Ⅲ: Extraction and electrophoretic analysis of ConABP in goat and cattle bileConABP was extracted from bile of goat and cattle by affinity chromatograph, and further analized by electrophoretic analysis. Results showed that proteins with four different molecular weight were included in ConABP (28.5Ku,24.4Ku,16.2Ku and 13.0Ku) in cattle bile, and the same in the goat bile. The straps of ConABP were at the low-middle area of track, and ConABP were four proteins with small moleculars weight.Ⅳ:The concentration change of ConABP,total protein,bilirubin and calcium-ion in bile during the formation of bezoar in the external calculus bovis generatorsThe E.coli(bacterial group) andβ-glucuronidase(enzyme group) was used to study the formation of bezoar in the external calculus bovis generators. Results indicated that the nucleation time(NT) of bacterial group was 3.7±0.2h, while the enzyme group was 0.75±0.2h. The nuclear density of bacterial group which increased gradually with the time increases slowly within 18 hours, while it increased at a relatively rapid speed between 18 hours and 68 hours, and peaked at 68 hours, then decreased gradually. The nuclear density of enzyme group which increased rapidly within 12 hours increased slowly between 12 hours and 18 hours, and peaked at 18h, then decreased gradually to stabilization. The nuclear size increased gradually with the time. The concentration of protein in bile appeared to have a decline trend after an initial ascent. The concentration of bilirubin decreased gradually with fluctuations. The concentration of Ca2+ decreased gradually mainly. The proportion of ConABP appeared to have a decline trend after an initial ascent, because it was mainly the high-molecular-weight protein participate in the formation of bezoar at the preliminary stage.Ⅴ: The concentration change of ConABP,total protein,bilirubin and calciumion in bile during the formation of bezoar in gallbladders of goatThe foreign object(foreign object group) and the E.coli(bacterial group) was implanted into gallbladders of goat to study the effect of the formation of bezoar. The nuclear density and size both appeared to have a decline trend after an initial ascent. The density of protein in bile (bacterial group) increased gradually within 12 hours, then decreased gradually to a stabilization above the initial value; and the density of bilirubin and Ca2+ both appeared to have a decline trend. The proportion of ConABP appeared to have a ascend trend within 24 hours, and then declined. The concentration of protein in bile (foreign object group) appeared to have a slowly ascending trend, while the concentration of bilirubin and Ca2+ both appeared to have a declineing trend; and the proportion of ConABP which decreased gradually within 24 hours appeared to have a ascending trend between 24 hours and 144 hours, and declined after 144 hours.Ⅵ:The pathologic changes of the goat gallbladder which foreign object was planted inThe gallbladder wall was extracted to study the pathologic change after the foreign object was planted. Fifteen days after the plantation, significant hyperplasy of mucous gland in lamina propria could be detected, and goblet cells are present in epithelium mucosae; the cell density of mucosal fold increased, and pseudostratified epithelium and nucleus squeeze was presented. Fifteen days after the plantation, the mucous gland hyperplasy continuely, and intestinal metaplasia and notho-glandulae pyloricae-metaplasia was present. Glandulae pyloricae similarity was seen in gallbladder wall.
