Extraction And Separation Of Flavonoid Contents In Abutilon Theophrasti Medic. And Its Anti-microbial Activity

In this thesis , the flavonoid contents in Abutilon theophrasti Medic. and its antimicrobial activity has been studied , And fatty acid composition of seed oil were studied by GC-MS method . the results are as follows :In this test, it was proved that flavonoid existed in Abutilon theophrasti Medic. every nutritious organs with qualitative analysis experiment . The optimum extraction condition of total flavonoids in Abutilon theophrasti Medic. stem was investigated by single factor selecting experiment and orthogonal experiment , The results as follows: methanol concentration was 70%,solid—liquid ratio was 1:40,refluxing temperature was 60℃and extracting duration was 1.5 hours. In this test Flavonoids in Abutilon theophrasti Medic . every nutritious organ was investigated by spectrometric method with rutin as standard material. The results showed that in Leaves have the highest concentration of flavonoids;flower and stem take the second place. The method is simple,convenient and can be applied to extract Abutilon theophrasti Medic .in large scale.In the test, 8 species of common food contaminating microorganisms were used as tested microorganism, and the antimicrobial activity of three solvents extracts of leaf of Abutilon theophrasti Medic. were studied by using the Beating holes method. The minimal inhibitory concentrations(MIC) were determined in this study.The results showed that the three kind solvent extracts could obviously restrain the growth of both bacteria and fungus at different digree .In the last part , Abutilon theophrasti Medic .seed oil was extracted by soxhlet method and its common physical-chemical properties were studied,and its fatty acids composition were analyzed by Gas chromatography–Mass spectrometry.The result showed that the extraction ratio is 19.07%.The refractive index of the seed oil is 1.47,the iodine value is 128.9gI/100g,the acid value is 8.04mg(KOH)/g,the saponification value is 266.47mg(KOH)/g。The 15 kind fatty acid are identified in Abutilon theophrasti Medic .seed oil,the main fatty acids are 9-Octadecenoic acid (25.57%),Palmitic acid(18.43%),linoleic acid (3.14%),9,12-Hexadecanoic acid (5.66%),Isostearic acid (4.8%),Stearic acid (1.54%),the total content of the unsaturated fatty acids is 87.4%.
