To find out an optimal extraction condition for Elaeagnus mollis alkaloids, the study compared with different extract solvents,different extract methods on the paste-forming rate. The results showed that, paste-forming rate was the hingest when acidic ethanol was used as solvent; Ultrasound extraction was the best extract menthods,the optimal extraction process for ultrasound extraction as follows: extraction for 3×25min with 85% ethanol as the extraction solvent, at 50℃, the rate of paste-forming was 0.44%.Studied the adsorption and desorption of five models of macroporous resin for E.mollis alkaloids, in order to separate and purification of E.mollis alkaloidsbetter. The results showed that, the type of D4020 is most beneficial to the separation and purify cation of E.mollis total alkaloids, and 70% ethanol was the best eluant.Two kinds of alkaloids get from the chloroform layer of non-phenol tertiary amine alkaloids, because of their volume were too low, failed to detect its structure.The total alkaloids were ultrasonic-assistedly extracted from Elaeagnus mollis, and were systematically separated into five groups. Then, their antimicrobial activities were assayed. Theresults showed that five alkaloids groups all have antimicrobial on the B.subtilis,S. aureus,V. ceratosperma and Verticillium dahliae,but no-obvious activity on the E.coli;In the five alkaloids groups, non-phenol tertiary amine alkaloids had the strongest antimicrobial activity, the MIC values of B.subtilis and S.aureus were 0.4mg/mL and 0.8mg/mL;when the density of non-phenol tertiary amine alkaloids was 3.2mg/mL, the inhibition activity of V. ceratosperma and Verticillium dahliae were 72.7% and 93.4%;The heat-resistant of Elaeagnus mollis's alkaloids was strong, after high temperature treatment did not reduce the activity of antibacterial; Decreased the pH value of antibacterial liquid by acid processing would reduce its antimicrobial activity, the acidification antibacterial liquid had the strongest antimicrobial activity When the pH value was 7.
