Objective:Determining the expression of MK, Ang-2and MVD with markers of CD105 in GIST tissues and analyzing the relationship between the expression of MK, Ang-2 and MVD, as well as clinical significance in GIST, whether or not can be used as non-specific target in diagnosis and assessment of the biological behavior of GIST. The survival analysis to explore the MK, Ang-2 and MVD with markers of CD105 with GIST prognosis that combination of follow-up data, its ability to predict prognosis of GIST patients with the independence of indicators;Methods:90 GIST patients whose pathological diagnosis was defined were selected from general surgery department of my hospital from January of the year 2001 to may of the year 2007 and 20 normal mucosa specimens. We took tissue microarray from paraffin blocks preserved in pathological department and determinate the protein expression of MK, Ang-2 and CD105 in it through immunohistochemical method(S-P method). In all cases were received follow-up data by telephone or letter. Experimental data were analyzed together with the clinical pathological datum.Result:1. The mean value of MVD with markers of CD105 was (6.01±1.78) and (3.48±1.02) in GIST and normal mucosa, respectively( p﹤0.01). The positive staining of MK and Ang-2 were mainly located in cytoplasm of tumor cells in GIST. The expression rates of MK and Ang-2 in GIST were 64.4% and 60.0% respectively, but there was no expression in normal mucosa( p﹤0.01) .2. The positive expressions of MK and Ang-2 and MVD were connected with tumor diameter, nuclear division rating and tumor risk of GIST( p﹤0.05或p﹤0.01), but had nothing to do with patients age, sex and initial occurring position of the tumor( p >0.05) in GIST.3. MVD was significantly higher in tumor tissue MK expression of positive than that expression of negative (6.32±3.29 vs. 4.19±1.51, p﹤0.05). MVD was significantly higher in tumor tissue Ang-2 expression of positive than that expression of negative (6.69±3.26vs. 4.07±1.53, p﹤0.05). MVD was significantly higher in tumor tissue both positive MK and Ang-2 than in that both negative MK and Ang-2 (7.07±3.26 vs. 4.02±1.91, p﹤0.05). The positive expression of MK and Ang-2 is correlated significantly(r =0.8105, p﹤0.05).4. Positive expression of MK, Ang-2 and high MVD were all associated with a poor survival about single factor analysis, but only MK was independent prognostic factor on multivariate analysis.Conclusion:1. The MVD with markers of CD105 and the positive expression of MK, Ang-2 in GIST were significantly higher than that of normal gastric mucosa, which may be relate to the occurrence and development of GIST.2. MK and Ang-2 promote angiogenesis, which are important to angiogenesis factor in GIST. They are relation to proliferation, invasion and metastasis, which can be used as non-specific target in diagnosis and assessment of the biological behavior of GIST.3. The expression of MK and Ang-2 is correlated significantly. There may be some coordination of them in regulating mechanism of tumor angiogenesis. MK and Ang-2 are involved in the process of the development of GIST. 4. The positive expression of MK, Ang-2 and high MVD with markers of CD105 are impact to prognosis of patients, which can be used as valuable markers of tumor prognosis for GIST.5. The expression of MK is independent prognostic factor in patients with GIST. It can be used as independence of indicator to evaluation the prognosis of patients with GIST.
