Objective:Our objective was to study the Magnetic resonance imaging (MRI) of nasopharyngeal carcinoma and its correlation with the expression of CD105 and cyclooxygenase-2 (COX-2) in nasopharyngeal carcinoma (NPC) and the microvessel density (MVD) stained by CD105 and CD34.Material and methods:78 case patients with nasopharyngeal carcinoma whose MRI was performed before treatment and the clinical staging had analyzed in relation to the clinical data were assessed. To analyse microvessel density and expression of CD105 and COX-2 was detected using immunohistochemical S-P method,and to study the relationship of those molecular markers, MVD and clinical staging.Result:1. The study of MRI showed T1 staging of NPC which localized in nasopharyngeal recess were found in 3 cases; T2,T3 and T4 staging of NPC were found in 35,24,and 16 cases, respectively.the retropharyngeal lymph node (RLN) and neck lymph node metastasis of NPC was found in 54 cases, the distant metastasis of NPC was found in 5 cases.2.The percentage of The CD105 and COX-2 positivity were 37.2% and 38.5%, respectively. The expression of CD105 and COX-2 was correlative with the different T staging, the groups with or without lymph node metastasis,and groups with or without distant metastasis of nasopharyngeal carcinoma (P<0.05 ) .The correlation between the expression of CD105,COX-2 and the groups with or without parapharyngeal space(PPS) involvement was no statistically significant (P>0.05) .3. The microvascular densities (MVD) of nasopharyngeal carcinoma stained by CD105 in T1T2,T3,T4 staging were 11.33±5.22,33.55±7.43, 56.71±9.64, respectively. The correlation between MVDcd105 and deifferent T staging present statistic significant (P<0.05). The correlation between MVDcd105 and the deifferent groups with or without lymph node metastasis,and groups with or without distant metastasis of nasopharyngeal carcinoma was also statistically significant(P<0.05). The correlation between MVDcd105 and the deifferent groups with or without parapharyngeal space involvement of nasopharyngeal carcinoma was no statistically significant(P>0.05). CD105 was not expressed in the vascular endothelial cells of any normal nasopharyngeal tissue but was expressed in the vascular endothelial cells of all nasopharyngeal carcinoma tissue. In contrast, CD34 was expressed in the vascular endothelial cells of normal nasopharyngeal tissue and nasopharyngeal carcinoma tissue.4. The correlation between MVDcd34 and deifferent T staging was no statistically significant (P>0.05) . The correlation between MVDcd34 and the deifferent groups with or without parapharyngeal space involvement, groups with or without lymph node metastasis,and groups with or without distant metastasis of nasopharyngeal carcinoma was also no statistically significant (P>0.05) .5. There was no significant correlation between positive CD105 and positive COX-2 expression of nasopharyngeal carcinoma. There was significant correlation between positive CD 105 expression and MVDCD105 of nasopharyngeal carcinoma.Conclusion:1.MRI are helpful imaging methods for accurate evaluate the local invasion, lymph node metastasis and distant metastasis of nasopharyngeal carcinoma.2.The present findings suggest the expression of CD105 and COX-2 may play an important role in the local invasion, lymph node metastasis and distant metastasis of nasopharyngeal carcinoma, which may serve as a potential clinical indicator for diagnose nasopharyngeal carcinoma.3.There was no significant correlation between CD105 and COX-2 expression of nasopharyngeal carcinoma.4.There was significant correlation between positive CD105 and MVDCD105 in nasopharyngeal carcinoma, the microvessel density (MVD) stained by CD105 was correlative with the different clinical staging, In contrast, the microvessel density (MVD) stained by CD34 was not correlative with the different clinical staging.CD105-MVD as compared with CD34-MVD is relevant a significant and independent prognostic indicator for tumorangiogenesis of nasopharyngeal carcinoma.
