Background: Colorectal carcinoma (CRC) is a kind of malignant tumor frequently occurred in gastrointestinal system and its morbidity has been rising in recent years all over the world. the incidence in China is also high,however,the mechanism about tumor immunology is not clear yet.Recently,people find that not only specific immune response but also nonspecific immune response play an important role in the antitumor immunology and immune surveillance by more study on the tumor immunological mechanism.It has been found in recent study that TLR is an important factor in nonspecific immune response and a bridge from nonspecific to specific immune.Toll-like receptors are a family of type I transmembrane receptors, they bound pathogen associated molecular patterns (PAMPS), through myeloiddiffer entiation factor 88 and nuclear factor, induce many signal pathways, then initiate cell activation, upregulate major histocompatibility complex (MHC) and costimulatory molecules expression,secrete cytokine and so on,at last,they induce inflammation response.At now,many researches have been tested that in TLR family,Toll-like receptor4 and toll-like receptor9 have a intimate relationship with tumor formation,but they whether have the same relationship with colorectal carcinoma still unknown,which need us to research.Objective: This study was designed to investigate the protein expression of TLR4 and TLR9 in colorectal carcinoma and tumor-free colorectal tissues,and analyse the relationship of TLR4 and TLR9 with various clinicopathological. To explore the relationship between expression of TLR4 and TLR9 and oncogenesis development and clinical characteristic of colorectal carcinoma.Methods: In this study, Immunohistochemical methods were used to investigate the expression of TLR4 and TLR9 in 40 cases with colorectal carcinoma and 20 cases with tumor-free colorectal tissue.Results:â‘ TLR4 and TLR9 positive stained were a greater or lesser degree detected in all of the 40 cases.Positive expression of TLR4 and TLR9 were found 67.5% (27/40) and 82.5% (33/40) in colorectal carcinoma, significantly higher than 10%(2/20) and 5% (1/20) in 20 cases tumor-free colorectal tissues evidently(P<0.05);â‘¡The expression of T LR4 and TLR9 in colorectal carcinoma had no relationship with patient's age, gender, tumor size, location (P>0.05);â‘¢The expression of T LR4 and TLR9 in colorectal carcinoma was closely related to the degree of differentiation, lymph node metastasis and Dukes stages(P< 0.05);â‘£there were close Compatibility among TLR4 and TLR9 in colorectal carcinoma (P< 0.05).Conclusions:â‘ Abnormality higher expression of TLR4 and TLR9 in colorectal carcinoma than tumor-free colorectal tissues;â‘¡The expression of TLR4 and TLR9 are associated with tumor's differentiation degree, lymph node metastasis and Dukes stages the more Tumor degree, Dukes stages and lymph node metastasis. the more expression of TLR4 and TLR9.this Prompted the two may be involved in the occurrence and the development process of colorectal carcinoma;â‘¢There were close Compatibility among TLR4 and TLR9,suggesting that the two may be synergies from the occurrence and development process of colorectal carcinoma;â‘£Be the participator who is nonspecific immunity, the high expression of TLR4 and TLR9 in colorectal carcinoma make the reagent which strengthen TLR4 and the TLR9's signal rotating leading the passage effect becoming fight very good tumor immunity assistant agent,then Applying TLR4 and TLR9 's ligand as immune adjuvants to treat colorectal carcinoma can become possible.
