Purpose To study the expression of EphA2 and E-cadherin in laryngeal squamous cell carcinoma(LSCC) ,and the relationship of the expression with the clinical pathology characteristics of LSCC ,and also to understand the correlation between the expression of EphA2 and E-cadherin in LSCC.Methods 62 samples of paraffin wax embed laryngeal squamous cell carcinoma were collected for the present study. These samples were cut off from surgical operations and stored in pathologic section of the people's hospital of Hunan province from 2002 to 2006. 60 samples of normal squamous epidermis near cancer were selected. The patients' age, sex, differentiation, cervical lymph node matastasis and TNM stage were recorded. All samples were recuted to diagnose. The pathological specimen was cut in the thickness of 4um. Streptavidin peroxidase (SP) immunohistochemistry staining for EphA2 and E-cadherin were made. The result of immunohistochemistry were determined by the intensity of staining and the number of positive cells. The expressions of EphA2 and E-cadherin in LSCC,and normal squamous epidermis near cancer were analyzed. The relationship with the expression of EphA2 and E-cadherin with the clinical pathology characteristics were done. and the relevance of the expression of EphA2 and E-cadherin in LSCC was analysed.Result1 The positive rate of EphA2 expression was 82.26%(51/62) in LSCC,and in normal squamous epidermis near cancer was 36.67%(22/60). The expression of EphA2 in LSCC was significantly higher than that in Lnormal squamous epidermis near cancer (P<0.01). 2 The positive rate of E-cadherin expression was 45.16%(28/62)in LSCC,and in normal squamous epidermis near cancer was 93.33%(56/60). The expression of E-cadherin in LSCC was significantly lower than that in Lnormal squamous epidermis near cancer (P<0.01).3 There was a significant correlation between the expression of EphA2 in LSCC with the type of pathology, cervical lymph node matastasis and the tumor TNM stage (P<0.05). However There was no significant correlation between the expression of EphA2 with the patients' age and sex (P>0.05) .4 There was a significant correlation between the expression of E-cadherin in LSCC with the type of pathology, cervical lymph node matastasis and the tumor TNM stage(P<0.01). while There was no significant correlation between the expression of E-cadherin with the patients' age and sex(P>0.05) .5 In LSCC, The expression of EphA2 was inversely correlated with that of E-cadherin(r=-0.325,P<0.01).Conclusion1 The expression of EphA2 was significantly increased in LSCC .Higher EphA2 expression was significantly associated with the type of pathology ,cervical lymph node matastasis and the tumor TNM stage, but not related with the patients' age and sex.EphA2 may accelerate the genesis or progress of LSCC. Higher EphA2 expression may imply more aggressive malignant behaviors of LSCC.2 The expression of E-cadherin was significantly reduced in LSCC.Dscending E-cadherin expression was significantly associated with the type of pathology ,cervical lymph node matastasis and the tumor TNM stage, but not related with the patients' age and sex. E-cadherin may inhibit the genesis or progress of LSCC. Dscending E-cadherin expression may imply more aggressive malignant behaviors of LSCC.3 In LSCC, The expression of EphA2 and E-cadherin were nagatively correlated.
