ObjectiveTo investigate the expression of Snail and E-cadherin, N-cadherin mRNA in laryngealsquamous cell carcinoma,and to discuss the relationship between the three enzymes andthe invasion and metastasis of laryngeal squamous cell carcinoma,and then to look forsome tumor markers which might prognose the invasion and metastasis of laryngealsquamous cell carcinoma to be used in clinical.MethodsTissue of LSCC were obtained from43patients who underwent surgery as forExperimental group, and according to2002TNM staging made by UICC,the tissues ofLSCC were divided into T1+T2group(26cases) and T3+T4group(17cases),as well asnon-metastasis group(LN-30cases) and metastasis group(LN+13cases).We collect another10cases adjacent tissues as control group.We detected the expression of Snail andE-cadherin, N-cadherin mRNA in every case by the technique of fluorescent quantitationRT-PCR. According to the statistical analysis of relative expression level of three proteinmRNA, we explore the relationship of possible correlation and clinical and pathologicalfeatures of laryngeal squamous cell carcinoma.Results(1)The relative expression of E-cadherin mRNA of laryngeal squamous cellcarcinoma tissues(2.19士0.72) was lower than that of adjacent tissues(28.17士0.98);GroupT3+T4(2.14士0.67) was lower than that of group T1+T2(2.92士0.51); Group LN+(1.73士0.61) was lower than that of group LN-(2.27士0.65). P<0.05,the difference wasstatistically significant.(2)The relative expression of N-cadherin mRNA of laryngeal squamous cell carcinoma tissues(26.35士0.82) was higher than that of adjacent tissues(1.81士0.45);Group T3+T4(29.40士0.98) was higher than that of group T1+T2(19.07士0.63); GroupLN+(31.13士0.81) was higher than that of group LN-(21.35士0.65).P<0.05,the differencewas statistically significant.(3) The relative expression of Snail mRNA of laryngeal squamous cell carcinomatissues (25.84士0.73) was higher than that of adjacent tissues (2.41士0.76);Group T3十T4(27.11士0.95) was higher than that of group Tl+T2(21.42士0.83); Group LN+(30.28士0.63) was higher than that of group LN-(23.18士0.54),P<0.05,the difference wasstatistically significant.(4)The expression of E-cadherin mRNA was correlated with tumor invasion, degreeof differentiation, clinical stage and lymph node metastasis (P<0.05), but not correlatedwith age, tumor size and tumor position(P>0.05) in laryngeal squamous cell carcinoma.(5) The expression of N-cadherin mRNA was correlated with tumor invasion, degreeof differentiation, clinical stage and lymph node metastasis (P<0.05), but not correlatedwith age, tumor size and tumor position(P>0.05) in laryngeal squamous cell carcinoma.(6) The expression of Snail mRNA was correlated with tumor invasion, differentiationdegree, clinical stage and lymph node metastasis (P<0.05), but not correlated with age,tumor size and tumor position(P>0.05) in laryngeal squamous cell carcinoma.(7) The expression of Snail mRNA was negatively correlated with the expression ofE-cadherin,but positively correlated with the expression of N-cadherin.And there was anegative correlation between mRNA expression of E-cadherin and N-cadherin.Conclusion(1)The expression of Snail and N-cadherin mRNA in laryngeal squamous cellcarcinomatissues upregulate,and the expression of E-cadherin downragulate,which mightbe of great value in the diagnosis of laryngeal squamous cell carcinoma.(2)The Snail and N-cadherin mRNAwere highly expressed in group T3+T4and groupLN+,these two proteins might promote the invasion and metastasis of laryngeal squamouscell careinoma,which might be of great value in the determination of therapeutie regimensand prognosis of laryngeal squamous cell careinoma.(3) The expression of E-cadherin mRNA were high in group T1+T2and groupLN-,which suggest that E-cadherin might play a role in the suppression of the invasion andmetastasis of laryngeal squamous cell carcinoma,and then pave away for finding a newantitumor drug. (4) The expression of Snail mRNA were negatively correlated with the expression ofE-cad mRNA, but positively negatively related to expression of N-cad mRNA,and theexpression of E-cad mRNA were negatively correlated with the expression of N-cadmRNA.It is suggested that snail may be involved in regulation of E-cad and N-cad bysome signalling pathways, and then affect the tumor invasion and metastasis.
