The Effect Of Chinese Medicine Qingyi Tang On TNF-α,IL-6,IL-10 In Rats With Acute Hemorrhagic Necrotizing Pancreatitis

Objective We observe the changer of the concentration on pro-inflammatory cytokines (TNF-α, IL-6) and anti-inflammatory cytokines (IL-10) in both serum and ascites and calculate the histopathology score of pancreas in rats with acute hemorrhagic necrotizing pancreatitis(AHNP) through building the model of AHNP rats and intragastric administration Chinese medicine QingYi Tang, to discuss preliminarily the mechanism of action of QingYi Tang on AHNP and to supply theory and experimental foundation for clinic.Methods Thirty SD rats were divided into three groups randomized (sortition): sham operated group(SO group, n=10), acute hemorrhagic necrotizing pancreatitis group (AHNP group, n=10), QingYi Tang group(QYT group, n=10). The SO group were be performed an operation of only flipping the pancreas. After opened the abdomen, the AHNP group and QYT group use a method of injecting 5% sodium taurocholate(0.1ml/100g) under the pancreas envelope to build the model of AHNP . After building the animal models and closed the abdomen, rats of the QYT group were be intragastriclly administered QingYi Tang(1ml/100g)at once and were be repeated one time 12 hours later. And rats of the SO group and AHNP group were just be intragastriclly administered normal sodium(1ml/100g). After modeled 24 hours later, rats of three groups were be opened the abdomen another time, to observe ascites, saponified spots and pancreatic gland through eyes. Then, we were determine the concentration of blood amylase, blood calcium, serum TNF-α,IL-6,IL-10 , ascites TNF-α,IL-6,IL-10 , and calculating the histopathology score of pancreas.Results (1)After modeling 24 hours, rats of the AHNP group were been opened the abdomen and it can be seen of hemorrhagic ascites, saponified spots, lamellar hemorrhage and necrosis on pancreas. And compared with the SO group, the blood amylase was higher(6131.9±1179.8 VS 175.4±59.9,P<0.01), the blood calcium was lower(1.7±0.2 VS 2.5±0.1,P<0.01), the histopathology score of pancreas was higher(9.8±1.0 V 1.4±2.8,P<0.01). (2)After modeling 24 hours, rats of the QYT group were been opened the abdomen and it can be seen of light hemorrhagic ascites, saponified spots, punctiform or lamellar hemorrhage and necrosis on pancreas. And compared with the SO group, the blood amylase was higher(1202.2±426.6 VS 175.4±59.9,P<0.01), the blood calcium was lower(2.2±0.2 VS 2.5±0.1,P<0.01), the histopathology score of pancreas was higher(8.2±1.6 V 1.4±2.8,P<0.01).(3)AHNP group had compared with SO group, the serum TNF-αhigher(62.7±7.3 VS 4.5±0.7,P<0.01),serum IL-6 higher(46±4.0 VS 11.3±0.7,P<0.01),serum IL-10 higher(11.2±0.4 VS 4.9±1.0,P<0.01). (4)Rats of the AHNP group, the concentration of TNF-α,IL-6,IL-10 in ascites were higher than the concentration in serum(1022.9±132.2 VS 62.7±7.3, P<0.01; 123.8±28.2 VS 46±4.0,P<0.01; 128.9±21.1 VS 11.2±0.4,P<0.01). (5)After intragastric administration QingYi Tang on the rats with AHNP(the QYT group), the concentration of TNF-α,IL-6,IL-10 in ascites and serum all cut down. Compared with the AHNP group, it has significant difference (P<0.01). (6) Compared with the AHNP group, the concentration of blood amylase in the QYT group were lower(1202.2±426.6 VS 6131.9±1179.8,P<0.01), the concentration of blood calcium were higher(2.2±0.2 VS 1.7±0.2,P<0.01), the histopathology score of pancreas were lower(8.2±1.6 VS 9.8±1.0,P<0.05).Conclusion (1)Rats of the AHNP group, the concentration of TNF-α, IL-6, IL-10 in serum all rose, which indicated a"high level balance"in course of acute hemorrhagic necrotizing of rats. (2)The concentration of TNF-α, IL-6, IL-10 in ascites of AHNP rats were higher than those in serum, which indicated it has important significance that we handle the pancreatitis associated ascitic fluid(PAAF) on clinic. (3) Compared with the AHNP group, the QYT group cut down the concentration of all of TNF-α, IL-6, IL-10 in both ascites and serum, and it indicates that QingYi Tang could lower the"high level balance"of pro-inflammatory cytokines and anti-inflammatory cytokines in acute hemorrhagic necrotizing rats.
