Background: ApolipoproteinA1(ApoA1) is the major apoprotein constituent of high-density-lipoprotein(HDL), and is the predominant protein that is associated with the HDL cholesterol (HDL-C) particle. Both HDL-C and ApoA1 are key components of the reverse cholesterol transport process and are inversely correlated with the risk of coronary athero- sclerosis disease(CAD). The relationships of ApoA1-75bp/+83bp poly- morphisms with lipids levels and the occurrence of atheromatous plaques in patients with type 2 diabetes mellitus (T2DM) remain unclear.Objective: The purpose of this study is to explore the correlation of ApoA1 gene -75bp/+83bp polymorphisms to dyslipidemia.Methods: The restriction fragment polymorphisms(RFLP) at two MspI sites in the 5'end at -75bp and +83bp of ApoA1 were detected using PCR in 150 healthy subjects,123 patients with type 2 diabetes and athero- matous plaques, and 107 patients with type 2 diabetes not atheromatisis.Result: The genotype frequencies of GG,GA,AA and CC,CT,TT of the ApoA1 in group A,B and C were 58.3%,27.5%,14.2%,90.2%,9.8%,0%;62.6%,32.5%,4.9%,73.2%,23.6%,3.2%;63.6%,29.0%,7.5%,89.7%,10.3%,0%.In the three groups,GG genotype in the -75bp was the most frequency. No differences were found in the frequencies of genotypes and rare A allele for -75bp(P>0.05).The frequency of rare T allele for +83bp in group B was significantly higher than group C(0.15&0.051,P<0.05).In group C, the subjects had significantly higher serum mean concentration of ApoB and LDL-C compared with group B(P<0.05).And the female subjects had significantly higher concentration of HDL-C and ApoA1(P<0.05).Conclusion: The association of ApoA1 gene - 75bp/+ 83bp poly- morphisms with dyslipidemia is evident in Chinese of Dalian.The AA and TT genotypes of ApoA1 is associated with hyperlipidemia.
