Experimental Research On Regutating The Expression Of EGF,HSP90α And TGF-β 2by Chinese Recipe HuaJiYiGanJian

Objective:To study the function of Chinese recipe HuaJiYiganjian on regulating the expression of Epidermal growth factor( EGF),Heart Shock Protein 90α(HSP90α) and Transforming Growth Factor-β2 (TGF-β2) in the transplanted hepatocarcinoma 22(H22)tumor of mice to discuss the possible mechanism of tumor resistance.Methods:Established the mice model of transplantation tumor of H22 carcinomatous.Divide into normal saline control groups,Chinese recipe group,5-Fu group and combined treatment group.Compared the inhibiting effect on tumor among the group after 14 days administration.The methods of immunohistochemical method and image pattern analysis were taken to quantitative detect and compare the expression of EGF,HSP90αand TGF-β2.Results:1.The experiment of every treated group resistance to the growth of transplanted hepatocarcinoma of mice showed:Tumor weight of control group(n=14) was 1.178±0.247g,while the tumor weight of HJYGJ group(n=12) was 0.657±0.132g,the resistant rate was 44.23%,it was obvio- usly lower than the control group(P<0.05).And tumor weight of 5-Fu group (n=14) was 0.757±0.268g,the resistant rate was 35.74%,it was obviously lower than the control group(P<0.05).And tumor weight of combined treatment group (n=13) was 0.532±0.074g,the resistant rate was 54.83%, it is obviously lower than the control group(P<0.05).2.Expression of EGF,HSP90αin transplanted hepatocarcinoma cells through immuno- histo- chemistry staining, our groups had positive expression.Image pattern analysis and one-factor analysis of variance showed that the expression of EGF,HSP90αimage in HJYGJ group was increased and was apparently different from that of the control group(P<0.05),while the expression of that in combined treatment group was also different from that of 5-Fu group.3.Expression of TGF-β2 in transplanted hepatocarcinoma cells through immunohistochemistry staining,Four groups had positive expression.Image pattern analysis and one-factor analysis of variance showed that the expression of TGF-β2 image in HJYGJ group was incr- eased and was apparently different from that of the control group (P<0.05), while the expression of that in combined treatment group was also different from that of 5-Fu group.Conclusion:HuaJiYiganjian has definite antitumor effect against trans- plantation tumor of H22 carcinomatous, the possible mechanism of its tumor-inhibiting could be correlated with inhibiting the expression EGF,HSP90αand promoting the expression of TGF-β2.
