Object:We designed the specific antisense oligonucleotides against one of the known human liver cancer related microRNA-224, to investigate the effects of antisense-microRNA-oligonucleotides-224 (AMO-miR-224) on the growth and apoptosis of liver related cell SMMC-7721. In addition, we wanted to explore its anticancer mechanisms.Methods:Selection of the effective concentration and time-course determination of the inhibitory effects of AMO-miR-224 on growth of cell line SMMC-7721.Flowcytometric analysis of cell apoptosis in cell line SMMC-7721 induced by AMO-miR-224. Real time PCR determination of microRNA-224 levels in SMMC-7721 cell line after incubated with AMO-miR-224.Result:At concentration of 0.3μmol/L and after incubation for 48h , AMO-miR-224 exhibited statistically significant inhibitory effects on proliferation of cell line SMMC-7721. This inhibitory effect of AMO- miR- 224 was observed stronger with incubation concentration. On the same condition, AMO-miR-224 promoted apoptosis of the cell line. The levels of microRNA-224 was decreased respectively by AMO-miR-224.Conclusions:The AMO-miR-224 against microRNA-224 can effici- ently inhibit growth and promot apoptosis of SMMC-7721 cell line, and Real time PCR demonstrated that AMO-miR-224 could decrease the level of microRNA-224 in SMMC-7721 cell line.
