The Study Of Promotion Of Saikosaponins-D On Apoptosis Of A549 Cells

Objective To study the mechanism of Saikosaponins-d restraint generation and promote apoptosis of A549 cells.Methods To detect inhibition of different concentrations of Saikosaponins-d on the A549 cells proliferation by MTT, to observe the apoptosis by TUNEL, to survey the level of bcl-2 and c-myc mRNA expression between before and after medication by RT-PCR, and to compare the Fas protein expression between before and after medication by Western blot.Results Saikosaponins-d play a role on promoting apoptosis of A549 cells, and there is positive correlation between apoptosis of A549 and dosage of Saikosaponins-d. The level of bcl-2 and c-myc mRNA expression decreased significantly after 48 hours.Conclusion Saikosaponins-d can restraint A549 cells to generate and promote A549 cells to apoptosis.
