Object: To study the effects of saikosaponins A and Baicalein on thecell proliferation and apoptosis in the human hepatocellular carcinoma cellline, HepG2cell line and SMMC-7721cell line.Methods: Through the experiment in vitro assays,saikosaponins A andBaicalein were used together to human hepatocellular carcinoma HepG2cells and SMMC-7721. The synergy effects to the proliferation of cells weretested by MTT method, the effect to the progress of cell cycle was tested byflow cytometry Propidium Iodide (PI) single st-andard method;the rate ofapoptosis was detected by flow cytometry Annexin V-FITC/PI doublelabeling method; and express level of cox-2and caspase-3protein of generelating to apoptosis was detected by Immunohistochemical method.Resμlts: In vitro cytotoxicity assay, the inhibition effect to the growthof HepG2and SMMC-7721cells produced by saikosaponins A、 Baicaleinand combination use of saikosaponins A and Baicalein. Flow cytometricdisplays48h after treatment with drugs, and compared to the control groupand two drug groups separately, apoptosis rate of HepG2and SMMC-7721cells produced by combination use of saikosaponins A and Baicalein aresignificantly increasing; flow cytometry cell cycle results show, incomparison with negative control groups, saikosaponins A and Baicalein onhepatocellμlar carcinoma HepG2cells and SMMC-7721after48h were ableto make significant increase in G0/G1cells, and make S, and G2/M periodcell significantly reduced,48h after treatment with the mixture of drμgsG0/G1period of HepG2cells and SMMC-7721cells number are increased,and have significant difference compared to the two drug groupsseparately,the results show that combination use of saikosaponins A andBaicalein have the synergy effect; immuno-cytochemical staining foundliver cancer apoptosis-related gene expression of COX-2lower andCaspase-3raise, and the effect of compatibility groups are clearly stronger than saikosaponins A group and Baicalein group.Conclusion: Our research demonstrated that: saikosaponins A andBaicalein can promote the rate of apoptosis and restrain the growth ofHepG2and SMMC-7721cells, the influence of compatibility groups canclearly bring synergistic reaction; The mechanism of the synergisticanti-tumor effect by saikosaponins A、 Baicalein and compatibility of twomay be due to regulate anti apoptosis died gene, and promoting apoptosisdied gene of expression and regulation cell cycle changes.
