| Backgrounds and Objective The dentritic cell is the strongest antigen presenting cell(APC) in vivo by now .During its maturation,it accomplishes the ingestion and submission of antigen,then activates na?ve T lymphocyte, produces cell toxic T lymphocyte(CTL) to start immune response ,which plays an important role in anti-tumor immune treatment.In the course of maturation, cell structure maybe have some corresponding changes to meet the cell function , which is our experimental stand.This experiment investigate the adaptive changes of ultrastructure of the dentritic cells(DCs) before and after the maturation .Methods To induce the murine bone marrow mononuclear cells into immatured dendritic cells with GM-CSF and IL-4,observing the cell morphology under the inverted phase contrast microscope and detecting the surface markers with FACS, then induce the imDCs into matured ones with LPS for 48 hours, observing the ultrastructures of DCs before and after maturation under transmission electron microscope and last make a comparative analysis . Results The dentritic processes of matured DCs after stimulated by LPS decreased significantly ,whereas the bioblast,ribosome and the diameter of nucleolus increased.Conclusion The results of the study illustrated that the distribution of surface processes maybe associated with the antigen-presenting capacity of DCs , and it might also be a potential ruler of cell function and status. |
