Objectives To investigate the protective effects of recombinant adeno-associated virus-adiponectin(rAAV2/1-Acrp30) on liver of experimental diabetic rats.Methods Fifty male healthy Wistar rats(180-220g) were randomly divided into two groups:normal control group(n=8,fed with ordinary diet) and the model group(n=42,fed with high-glucose and high-fat diet). Four weeks later,STZ(35mg/kg) was injected intraperitoneally to establish experimental diabetic rat models.Twenty-seven rats became diabetes and randomly divided into three groups:(1) diabetes control group(n=9);(2) adiponectin treatment group(n=9),injected (rAAV2/1-Acrp30) 1×1012 v.g/ml to hind limb intramuscularly;(3) vacuity virus group(n=9),injected recombinant adeno-associated viruses 2/1 to hind limb intramuscularly.The normal control group and the diabetes control group were injected intramusculary with equal volume of saline.Blood glucose,serum concentration of HDL,LDL,TC,TG,APN, and liver histology,liver expression of NF-κBp65,IκBα,IL-1βwere assayed after 4 weeks' treatment.Results(1) After high-glucose and high-fat diet for 4weeks and injection of STZ,the blood glucose and water consumption increased significantly(P<0.01),serum APN decreased significantly(P<0.01).(2) Compared with the diabetes control group and the vacuity virus group,the FBG and water consumption in adiponectin treatment group decreased significantly(P<0.05),serum TC,TG,LDL also decreased significantly(P<0.05),HDL increased significantly(P<0.05),while serum APN did not increased significantly(P>0.05).(3) Compared with the diabetes control group and the vacuity virus group,the structure of liver,fatty degeneration of liver tissues and hepatic inflammation in adiponectin treatment group markedly reduced.(4) Compared with the diabetes control group and the vacuity virus group,the expression of APN in adiponectin treatment group increased significantly(P<0.01),and its liver expression of NF-κBp65,IκBα,IL-1βdecreased significantly(P<0.05).Conclusions(1) rAAV2/1-Acrp30 can decrease blood glucose and blood lipids,improve the symptoms of diabetes,it suggests that adiponectin has alleviate function to blood glucose and blood lipids of diabetes mellitus.(2) rAAV2/1-Acrp30 can improve the liver inflammatory cells infiltrating and steatosis of hepatic cells,It suggests that adiponectin has protective effects on livers of diabetic rats.(3) The protective effects of rAAV2/1-Acrp30 may be produced by decreasing liver expression of NF-κBp65,IκBαand IL-1β.
