Nasopharyngeal carcinoma is one of the most common cancers in China,especially in Provinces of Guangdong, GuangXi, HuNan, HuJian. Polygene, multifactor and multistage are involved in the occurrence of NPC. The variations of gene level lead to a mount of secretory proteins, which is released to extra-cytolemma,including extracellular ground substance,growth factor, cytokine, peptide hormone, enzymes in plasma and tissue fluid. Secretory proteins play a great role in intercell information transfer,having a biologicallphysiology or pathological effect in a way of paracrine or autocrine. There are many protein molecules related with the carcinogenesis of NPC,which exist can induce directly the occurrence of carcinoma and keep their heteromorphism, and related with the invision and matastasis of carcinoma. They may work as tumor markers diagnose. We use proteomic techniques to separate and assess nasopharyngeal secretions with NPC,and use Immunohistochemistry to detect the differential expression of AnnexinA5 protein in NPC tissues.Objective:To establish 2-DE proteome profile of nasopharyngeal secretion of patients with nasopharyngeal carcinoma and healthy people by two-dimensional polyacrlamide gel electrophoresis(2-DE).To search for different proteins serving as markers of nasopharyngeal carcinoma in aspects of early diagnosis and prognosis. And search for the relevance of AnnexinA5 and carcinogenesis with NPC. Methods: To collect nasal lavage fluid from patients with nasopharyngeal carcinoma and healthy people,and the homogeneous samples were mixed. The proteins were separated by 2-DE with PDquest software to analyze the differential expressions of proteins in 2-DE gels. Thirty four cases of nasopharyngeal carcinoma tissues and ten cases of chronic nasopharyngitis tissues were collected,formalin-embeded and paraffin-fixed,then they were stained using the method of streptavidin-peroxidase(SP) immunohistochemistry, then were analyzed with computed image analyzer and SPSS 11.5 software to find out differential expressions of AnnexinA5 protein between the two groups,and to search for the relevance between AnnexinA5 and carcinogenesis of nasopharyngeal carcinoma.Results:Eleven different protein spots were screened between the NPC and healhy control group,of which three protein spots showed up-regulation,eight protein spots showed down-regulation. The AnnexinA5 protein expression was significantly higher in NPC than that of nasopharyngitis(P<0.05),There was no significant relationship between the expression of AnnexinA5 protein and lymph node metastasis with NPC.Conclusion: Through the differentiated analysis of these nasopharyngeal secretion 2-DE gels,the differences of secretion proteome profiles,including numbers and densities,were discovered between the NPC and healthy control groups. And some special secretory Proteins maybe related to mechanism of nasopharyngeal carcinoma. The tumour molecular marker AnnexinA5 maybe related to the occurrence of NPC.We got a new clue and method for further research of carcinogenesis of nasopharyngeal carcinoma...
