ObjectiveTo detect the expression of SATB1mRNA in5-8F、6-10B、NP69-SV40T celllines and the expression of SATB1protein in nasopharyngeal carcinoma(NPC) tissue,and to investigate the clinical value of SATB1in NPC.MethodsThe expression of SATB1mRNA were investigated in5-8F、6-10B andNP69-SV40T cell lines by quantitative real-time PCR(qRT-PCR);Immunohistochemical staining method was used to detect the expression of SATB1protein in72cases of NPC and30cases of chronic nasopharyngitis tissue(CNT), andthe correlations between SATB1expression and clinicopathological parameters wereevaluated.Results1.FQ-RT-PCR for quantification of SATB1mRNA was successfully established.The efficiency of PCR is above90%. The expression of SATB1mRNA in5-8F cellline was lower than that in NP69-SV40T cell line, but the difference was notstatistically significant(P>0.05); The expression of SATB1mRNA in5-8F cell linewas4.09times higher than that in6-10B cell line(P<0.05).2.The expression of SATB1positive particles was manly localized in cell nucleus.Positive rates of SATB1protein expression in NPC and CNT were52.8%(38/72) and13.3%(4/30), respectively. The positive rate of SATB1in NPC cases were significantly higher than that in CNT(P<0.05). Expression intensity of SATB1inNPC was positively associated with lymph node involvement and distant metastasis(P<0.05). The up-regulated expression of SATB1in NPC was not associated withgender, age, T stage and clinical stage(P>0.05), but it had positive relationship withN and M stage(P<0.05).Conclusions1.The expression of SATB1mRNA in5-8F cell line was4.09-fold higher thanthat in6-10B cell line(P<0.05), indicate that the expression of SATB1mRNA maybe associated with NPC invasion and metastasis.2.The positive expression of SATB1protein in NPC cases were significantlyhigher than that in CNT, prompting that SATB1may play an critical role in NPCtumorigenesis and development.3.The aberrant expression of SATB1protein may be involved in the invasion andmetastasis of NPC. ObjectiveTo study the effect and significance of cisplatin and X-ray on the expression ofSATB1gene in5-8F cell line.MethodsSurvival rate of5-8F cell with treatment of different concentration andtime-point of cisplatin was measured using MTT metabolic viability assay. Expressionlevel of SATB1mRNA in5-8F cell was detected by real time RT-PCR after treatedwith cisplatin and X-ray.ResultsThe proliferation of5-8F cell was inhibited in a dose-and time-dependentmanner after the treatment of cisplatin. After treated with cisplatin and X-ray, theexpression level of SATB1mRNA were up-regulated(P<0.05), but it had not dose-and time-dependent manner with SATB1mRNA expression(P>0.05).ConclusionsCisplatin and X-ray can up-regulate the expression of SATB1mRNA in5-8F cell.These date suggest that SATB1gene may be play an important role in drug resistanceand radiation resistance of NPC.
