Object To prepare PEG-PEI Fe3O4 nano-magnetic fluid and invest its characteristics.Mothods PEG-PEI Fe3O4 nano-magnetic fluid was prepared by chemical co-deposition as gene delivery vector.Then DNA of plasmid pAFP-CD/TK was wrapped into PEG-PEI Fe3O4 nano-magnetic fluid.At the same time,we invest its characteristics.Results PEG-PEI Fe3O4 nano-magnetic fluid -DNA complexes average diameter is 202.6nm,uniformly distributed,no visible cytotoxicity,the highest combination rate with DNA is 93.56%.It can protect the DNA of plasmid,and the exogenous gene expression is normal.AFP positive cells transfected by pAFP-CD/TK were highly sensitive to the GCV treatment,the survival of cells declined obviously. pAFP-CD/TK suicide gene therapy system had ten times sensitivity than the CD group and TK group in AFP positive HCC.Conclusion This new target strategy of pAFP-CD/TK suicide gene therapy system in which PEG-PEI Fe3O4 nano-magnetic fluid are used as gene delivery vectors explores a promising area for AFP positive HCC and associated carcinoma therapy,and the effect of CD/TK group with GCV/5-Fc are better than the CD group and TK group.
