Objective:To study the possible mechanism of BushenhuayuFang based on the the role of tonifying kidney and dissolving blood stasis treating female the sterility due to anovulation and study the intervention role of promoting the growth of follicle and ovulation.Methods:Androgen Induced Sterile Rats model(ASR) was established by injecting testosterone propionate subcutaneously to SD female rats of 9 days age.The sex cycles and Morphological change of ASR's ovary and Insulin-like growth factor binding protein-1 and Insulin-like growth factor binding protein-3 expressions at its ovary after feeding Chinese herbs extract of tonifying Kidney and dissolving blood stasis were observed by light microscope and in stitu hybridization.Results:1,BushenhuayuFang can induce the sex cycles of rats and increased the ovulation rate of ASR rats which greatly increased the weight of rats' ovary and the total of Ovum bubble quantity of different levels in ovary.2,BushenhuayuFang could strengthen the expression of the insulin-like growth factor binding protein-1mRNA in rats' ovary Insulin-like growth factor binding protein-1mRNA expressions and number of positive cells in rats' ovary of the model group was were lower than that of the normal group(P<0.05);Insulin-like growth factor binding protein-1mRNA expressions and number of positive cells in rats' ovary of the BushenhuayuFang treated group and the clomiphene(CC) treated group were higher than that of the model group(P<0.05);Insulin-like growth factor binding protein-1mRNA expressions and number of positive cells in rats' ovary of the BushenhuayuFang treated group and the CC treated group was lower to the normal group(P<0.05),Insulin-like growth factor binding protein-1mRNA expressions in rats' ovary of the BushenhuayuFang treated group were higher than that of the CC treated group(P<0.05)3.BushenhuayuFang could reduce the expression of the insulin -like growth factor binding protein-3mRNA in rats' ovary.Insulin-like growth factor binding protein-3mRNA expressions and number of positive cells in rats' ovary of the model group was were higher than that of the normal group(P<0.05);Insulin-like growth factor binding protein-3mRNA expressions and number of positive cells in rats' ovary of the BushenhuayuFang treated group and the CC treated group were lower than that of the model group(P<0.05);Insulin-like growth factor binding protein-3mRNA expressions and number of positive cells in rats' ovary of the BushenhuayuFang treated group and the CC treated group was higher than that of the normal group(P<0.05);but no ststistical difference was found among the BushenhuayuFang treated group and the CC treated group(P>0.05).Conclusion:BushenhuayuFang based on tonifying kidney and dissolving blood stasis can induce the sex cycles of ASR rats,improve its ovulation rate,greatly increase the weight of rats' ovary and the quantities of ovarian follicle of different levels in ovary.It facilitated the growth and ovulation of ovarian follicle which may be achieved through the adjustment of the function of hypothalamus-pituitary-ovary axis(HPOA). BushenhuayuFang can enhance the expression of IGFBP-1mRNA and decrease the expression of IGFBP-3mRNA in part of ASR rats' ovary,so to adjust biological activity of IGFs with mitogenesis which can facilitate the growth and ovulation of ovarian follicle,it may be one of its mechanisam of action.From that point of view,It proved that BushenhuayuFang based on the the role of tonifying kidney and dissolving blood stasis bears theoretical foundation on molecular research level to the treatment of Sterility of kidney deficiency and blood stasis and to the substaintial study of disease and syndrome of it.And further confimed that the facilitation to the ovulation of ASR by BushenhuayuFang is possibly realized by the manner of multi-level and multi-target.
