Objective The intestinal mucosal barrier is susceptible to ischemic injury.Low blood perfusion during stress processes for example small bowel transplantation,trauma,serious infection and hemorrhagic shock can result in Ischemia/Reperfusion Injury.Glutamine is considered as "conditionally essential amino acid" and acts as one of the most important metabolize energy sourses in human body.Gln is excessively consumed during aforementioned etiological processes.Recent studies show compensation of its excessive consumption and its important role on protecting mucosal barrier and preventing bacterial translocation by exogenous glutamine supply.In this paper,small bowel IRI rat models were established to explore the effection of glutamine enriched parenteral nutrition on mucosal barrier and the HO-1 expression,then discuss the probable mechanisms.Material and Method1.Animals and Experimental groups:Thirty Wistar rats were randomly divided into 3 groups:The control group(N group,n=10) conducted fictitious operation and fed with common forage,TPN group(n=10) and TPN+Glu group(n=10).2.Animal models:Expose the superior mesenteric artery(SMA) through the median incision afer 5 days adaptive feeding.No traumatic procedures were done on the rats of the control group,but the others'SMA were clamped for 30 minutes and reperfused for 4 hours.All the rats were intubated in external jugular veins to infuse parenteral nutrition.5 days later,all the rats were killed to get the experimental materials. 3.Observative index:the morphology of mucosus,serum and intestinal mucosal Gln concentration,levels of D-lactate,endotoxin and TNF-α,IL-6.HO-1 positive ratio was detected by immunohistological staining and HO-1mRNA expression by RT-PCR.4.Statistical analysis:Data were expressed as x±s.Statistical analyses of the data between comparasive teams were performed using t test.p<0.05 presents significant difference,p<0.01 presents greatly significant difference.Results:1.Exgenous glutamine supply obviously enhanced the level of serum and intestinal mucosal Gln concentration.2.Exgenous glutamine supply obviously ameliorated the mucosal morphology and decrease levels of D-lactate,endotoxin and IL-6.HO-1 positive ratio and HO-1mRNA expression were enhanced.Conclusion1.Exgenous glutamine supply can alleviate small intestinal IRI and inflammatory reaction,protect mucosal barrier.2.Exgenous glutamine supply can enhance the HO-1 and HO-1mRNA expression.HO-1 and its metabolic products have antioxidant,apoptotic and anti-inflammatory functions.
