Objective:To investigate the effect of Ginseengsaponin on local immune microenvironment in patients with hepatocellular carcinoma after transcatheter hepaticarterial chemoembolization. Animal experiments will explore the effect of ginseengsaponin on local immune microenvironment of H22 tumor-bearing mouse,provide more theoretical basis for clinical trials.Methods:Forty patients with hepatocellular carcinoma were randomly divided into two groups:the treatment group(n=20) and the control group(n=20).The patients in the treatment group were treated with TACE plus Ginseengsaponin before and after TACE,while those in the control group were treated only with TACE.The response rate,AFP value,local immune function,liver function,Karnofsky score,quality of life and adverse effect of all patients were measured before and after TACE therapy.Animal experiments are as cinobufotalin for the positive control,then observed the effect of ginseengsaponin on local immune microenvironment of H22 tumor-bearing mouse.Results:After therapy CD3+,CD4+,CD4+/CD8+,NK cell,IL-2 levels of treatment group were significantly higher than those of control group.Cinobufotalin,ginseengsaponin both can effectively inhibite the tumor growth of H22 tumor-bearing mouse.Conclusion:Clinical trials prompted ginseengsaponin can significantly enhance the immune function,relieve symptoms and reduce the adverse effects of interventional therapy; Experimental study showed that ginseengsaponin alone with inhibition rate in cinobufotalin no significant difference,but the combination can significantly improve on the H22 tumor-bearing mice inhibited tumor growth.
