Theoretical ResearchTo elucidate the disequilibrium of neurotransmitters(monoamine and amino acids) by conjuncting the traditional Chinese medicine and modern medicine.Clinical ResearchObjective:To study the effect of monoamine and amino acids neurotransmitter in the mechanism of MT.Methods:The levels of monoamine and amine acides neurotransmitters in MT group which has 10 MT children,were compared with normal group which has 10 healthy children. High performance liquid chromatography(HPLC) was applied to test the plasma levels of dopamine(DA),5-hydroxytryptamine(5-HT),norepinephrine(NE) and homovanillic acid (HVA),which is the metabolite of DA,and glutamic acid(GLU),aspartic acid(ASP),gamma aminobutyric acid(GABA) in the blood serum.Results:The plasma levels of 5-HT and the blood serum of GLU,ASP in MT group was higher than that of the normal group(P<0.05).But there was no significant difference in the plasma levels of DA,NE,HVA and the blood serum of GABA in the 2 groups(P>0.05).Conclusion:There is the disequilibrium of neurotransmitters,such as monoamine and amino acids,in MT children.Expirical ResearchObjective:To elucidate the function and mechanism of Jing An oral liquid for the treatment of Multiple tics(MT).Methods:40 SD mice were randomly divided into 4 groups,namely,the normal control group,model group,Jing An group(treated by Jing An liquid),and positive control group(treated by Tiapride).10 animals in each group. To develop a mouse model to simulate the behavioral and neurochemical changes of Multiple tics by DOI induction and to observe the stereotyped action on head twitch response(HTR) induced by DOI. The brain tissue and blood levels of dopamine(DA),5-hydroxytryptamine(5-HT),norepinephrine(NE)and homovanillic acid(HVA),which is the metabolite of DA,and glutamic acid(GLU),aspartic acid(ASP),gamma aminobutyric acid(GABA) were measured by high performance liquid chromatography(HPLC).Results:1.The brain tissue levels of DA,5-HT,NE in model group was significantly increased,compared with that of the normal control group(P<0.01).But there was no significant difference in the levels of HVA in 4 groups.2.Compared with that of the control group,the plasma levels of NE in the model group were decreased(P<0.05) and the plasma of DA and 5-HT were no significant difference(P>0.05).The result of HVA in the plasma was not founded. 3.There was no obviouse difference in the brain tissue levels of GLU,ASP, GABA,when compared with that of the normal control group(P>0.05).4.The blood seurm levels of GLU in model group was higher than that of the normal control group(P<0.05) and the result of ASP and GABA in the blood serum was not founded. Conclusion:1.The numbers of tic and stereotypic behavior were obviously reduced by Jing An oral liquid.2.The mechanism of treating effects may be related with the hyperfunction of dopaminergic system and the exitotoxicity of excitatory amino acids(EAA) to nerve cell were inhitited by Jing An oral liquid.3.The disequilibrium in the central and peripheral transmitters were existed in the moddle group induced by DOI.4.Mild drowsy,one of Tiapride's adverse reaction ,may be associated with the increased enormously level of the inhibitory amino acids GABA in the brain introduced by Tiapride.
