The Effect Of 654-â…¡ To The Positive Rate Of Proliferating Cell Nuclear Antigen In Colonal Smooth Muscle Of Scalded Rat

Objective:To explore presumed alteration in the positive rate of proliferating cell nuclear antigen in colonal smooth muscle of scalded rat with 654-Ⅱinjected in.Methods:120 healthy Sprague-Dawley rats(male,weight 250±30g) were randomly divided into four groups,30 per group.Group A for control,Group B for rat with physiological saline and 654-Ⅱinjected,Group C for scalded rats with physiological saline injected,Group D for scalded rats with physiological saline and 654-Ⅱinjected.12h before the start of the experiment,food was removed from the rats,but water is free to all of them.With dorsal hair removed and anesthetized,an approximately 30% TBSA third degree scald injury is made to the rat in Group C and Group D with boiling water for 15 seconds.After that,Group A and Group C was injected in physiological saline(50ml/Kg) in peritoneal cavity,Group B and Group D was injected with physiological saline(50ml/Kg) and 654-Ⅱ(1ml) in peritoneal cavity.These rats whose colonic smooth muscle was collected at 16h and 24h after scalding was injected in in peritoneal cavity with 654-Ⅱ(1ml) after scalding.Five rats` colonic smooth muscle was collected in each time(1h,3h,6h,12h,16h,24h) per group.Then the smooth muscle were digested to single cell suspension.At last these samples was taken to detect the positive rate of PCNA.Results:The consequence tell us that there is no difference in the positive rate of PCNA in both Group A and Group B,P>0.05.But we saw a clear difference in between Group A and Group C,P<0.05.At 1h,3h,6h,12h,the positive rate of PCNA in Group A is higher than Group C,but at 16h,24h the result is the opposite,P<0.05.All numbers in Group A is lower than the numbers Group D and the difference is obviously,P<0.05 .And the positive rate of PCNA in Group D is higher than Group C in 1h,3h,6h,12h,16h,but at 24h the result is the opposite too,P<0.05.Conclusion:The positive rate in Group C is lower than Group D in 1h,3h,6h,12h,16h,but in 24h the result is positive.It tell US that 654-Ⅱcould not only cure the injury of colonic smooth muscle in scalded rat,but also reduce the lesion.
