Objective To investgate the effect of recombinant human growth hormone(rhGH) on growth and migration of gastric carcinoma BGC-823 cells in vitro.Methods The human gastric carcinoma BGC-823 cells in logarithm growing period were cultured with variant concentrations of rhGH or rhGH combinedand L-OHP in RPMI 1640 medium supplemented with 10% fetal bovine serum (FBS) .The study included six groups:①NS group;②L-OHP(4μg/L) group;③rhGH1(100μg/L)group;④rhGH2 ( 200μg/L) group;⑤r hGH1 (100μg/L)+L-OHP(4μg/L)group;⑥rhGH2 (200μg/L)+L-OHP(4μg/L) group. The expression of GHR in BGC-823 cells was detected by using S-P immunohistochemistry techniques. Cell growth inhibition rates and growth curves and doubling time was determined by spectrophotometer assay (MTT) and cell counting. Transwell chamber model assay was used to investigate the effects of rhGH on migration in human BGC-823 cells line.Results (1) The Results of immunochemical assay showed that GHR weakly expressed in BGC-823cells and located at the cell's membrane and in the plasma and taken on brown particle. (2) According to the growth curve determined by MTT and cell counting, rhGH did not have effect on multiplication of BGC-823 cells, there was no distinct difference(P>0.05), compared with NS group. The growth inhibition rates of L-OHP and rhGH + L-OHP groups increased obviously(P<0.01),compared with NS and rhGH groups.There was no distinct difference between rhGH + L-OHP grouuPs and L-OHP group(P>0.05).The doubling time of BGC-823 cells were NS group(31.06±2.53h), L-OHP group(105.27±5.16h), rhGH1group(31.37±1.89h), rhGH2 group(31.38±1.26h),rhGH1+L-OHP group(107.04±5.96h),rhGH2+L-OHP group(99.38±5.01h), respectively. There was no obviously difference in cell doubling time between rhGH groups and NS group(P>0.05) The cell doubling time of L-OHP group decreased obviously(P<0.01), compared with NS and rhGH groups. There was no difference between rhGH+L-OHP groups and L-OHP group(P>0.05). (3) The result of Transwell chambers assay showed that the morphology of BGC-823 cells changed after penetrating the membrane,and that there was no obviously difference in number of penetrating membrane between rhGH groups and NS group(P>0.05). There was no difference between rhGH1 group and rhGH2 group (P=0.429).The number of migration cells per visual field were NS group(25.64±1.81),L-OHP group(9.00±0.90),rhGH1 group(26.50±1.91),rhGH2 group ( 27.17±2.34 ) ,rhGH1+L-OHPgroup ( 9.87±1.42 ) ,rhGH2+L-OHP group(10.63±1.91), respectively. The number of L-OHP group decreased obviously (P<0.01), compared with NS and rhGH groups. There was no difference between rhGH+L-OHP groups and L-OHP group(P>0.05).Conclusion 1. GHR was weakly expressed in BGC-823 cells.2. rhGH does not have effect on the growth and migration of BGC-823 cells.3. The inhibitied effect of L-OHP on the BGC-823 cells could not be significantly influenced by rhGH.
