Effects Of Recombinant Human Growth Hormone (rhGH) On GH,IGF-Ⅰ,IGFBP And VEGF In The Bearing Gastric Carcinoma Nude Mice's Blood Serum

Objective:To study effects on growth hormone(GH),insulin-like growth factor-Ⅰ(IGF-Ⅰ),insulin-like growth factor binding protein-1,2,3(IGFBP-1,2,3)and vascular endothelial growth factor(VEGF)in bearing gastric carcinoma nude mice's blood serum and effects of the multiplication of human gastric carcinoma cells in short-term applicating recombinant human growth hormone(rhGH).Methods:After cultured in vitro human gastric carcinoma cells MKN45 were inoculated in right buttock of nude mouse and carcinoma xenograft model was formed.The better inoculated tumor nude mice were selected after cells were inoculated in 14 days later.Mice were divided into 4 groups randomially:control group,cisplatin(DDP)group,rhGH group and DDP+rhGH group.The drugs were continuously administered for 6 days and after the drugs were administered,each group was divided into two time-spots(the first day and the third day).There were 6 nude mice in each group.After the experiment was finished,the eyeball of mouse was picked to draw blood on the first day and the third day,respectively.The serum level of GH,IGF-Ⅰ,IGFBP-1,2,3and VEGF were examined by the method of ELISA.IN the end,the long and short diameter of the tumor was measured and the tumor volume and inhibitory rate were computed.Results:Tumor:The tumor obviously grew slowly,the volume obviously reduced and the tumor inhibitory rate increased in DDP group and DDP+rhGH group,compared with rhGH group and control group(P＜0.05).However,there was no statistical significance between DDP group and DDP+rhGH group or between rhGH group and Control group(P＞0.05).Serology:On the next day of completing use of drugs later,GH,IGF-Ⅰ,IGFBP-2,IGFBP-3 of the blood serum increased obviously in the rhGH group and DDP+rhGH group,compared with the DDP group and control group(P＜0.05).However,therewas no statistical significance between rhGH group and DDP+rhGH group or between DDP group and control group(P＞0.05).IGFBP-1 of the blood serum decreased obviously in the DDP+rhGH group and DDP group, compared with the rhGH group and control group(P＜0.05).VEGF were decreased obviously in the blood serum of the DDP group,compared with the rhGH group and control group(P＜0.05).VEGF of the blood serum decreased obviously in the DDP+rhGH group,compared with the rhGH group(P＜0.05).However,there was no statistical significance between rhGH group and control group about IGFBP-1 and VEGF(P＞0.05).There were no statistical significance between the DDP group and DDP+rhGH group about IGFBP-1 and VEGF(P＞0.05)either.On the third day of completing use of drugs later,there were no statistical significance in GH,IGF-Ⅰ,IGFBP-1,IGFBP-2,IGFBP-3 among all of four group(P＞0.05).But VEGF decreased obviously in the DDP group and DDP+rhGH group,compared with the rhGH group and control group(P＜0.05).However,there was no statistical significance between rhGH group and control group about VEGF(P＞0.05).There were no statistical significance between the DDP group and DDP+rhGH group about VEGF(P＞0.05)either.Conclusion:GH,IGF-Ⅰ,IGFBP-2 and IGFBP-3 of blood serum of nude mouse bearing gastric carcinoma were increased aRer application of recombinant human growth hormone(rhGH)in short term,but the growth of human gastric carcinoma cells were not promoted.
