Objective:to discuss the effect of repairing bone defect in rabbits with PRP and BMP and PLGA.Methods:32 Japanese white rabbits were divided into three groups at random.Preoperative, BMP was combinated with PLGA carrier in the underpressure,PRP was prepared in the two procedure by centrifugal machine.After the 15mm defect experiment model in the right radius or rabbit was made,the rabbits were devided into three groups:BMP with PLGA carrier and PRP was planted in the experimental group,BMP with PLGA carrier was planted in the control group,and the blank group was handled with nothing.After 4,8,12 weeks, investigation of the bone defect was made by means of X-ray examination,gross observation, histology of HE staining.The score of X-ray examination was according to lane standard.Results:(1) Imageology analysis from the 4th week to 12th week,X-ray showed that the bone defects of the blank group were not repaired.4 weeks after operation,Both experimental group and control group showed up the continuity,more uniform density shadow,fracture lines to be fuzzy,the area of new bone in experimental group was more than in control.8 weeks after operation,there were more new bone increasing in experimental group and control group,fracture lines to be identified unclear,most region of bone defect to be found callus formation,density was close to the normal host bone.the area and density of callus formation in experimental group was slightly higher than in control.12 weeks after operation, the bone defects of experimental group and control group were repaired,continuous callus, new bone shaping good,and there formed a medullary cavity,the diameter of new bone of experimental group is slightly rougher than the normal host bone,in the control group is slightly smaller than the normal host bone,the shaping of bone defects of the experimental group were more superior than the control group.According to lane standard,the score of X-ray examination of experimental group was higher than control group in each period of time,there is statistical difference between experimental group and control group(P<0.05). (2) Gross observation showed,12 weeks after operation,the defects of both experimental group and control group had been repaired,the shaping of new bone in experimental group was better than in control,the diameter of new bone of experimental group is slightly rougher than the normal host bone,the bone defects of the blank group had not been repaired.(3) Histology showed,12 weeks after operation,the bone defects of both experimental group and control group had been repaired,the quantity and quality of new bone formation of experimental group was more higher than control group,and there was only fibration in the blank group.Conclusions:(1) The compound of PRP and BMP and PLGA for the treatment of bone defect is an effective,economic,convenient,low-injury treatment.(2) PRP can promote bone defect repairing and wound healing.
