Objective: Discuss the association of schizophrenia with genetic polymorphism presented by 15 Short Tandem Repeats loci.Methods: Fifty hospitalized patients in Dalian No. 7 People's Hospital (special hospital in mental diseases), including 25 males and 25 females averagely aged 46, constitute the observation group. The diagnosis standard was referred to U.S. Statistical Handbook for Mental Disease Diagnosis (fourth edition). One hundred and fifty blood samples were collected from 75 males and 75 females who had no obvious mental and body diseases from the Dalian Blood Center as the control group. Samples were anticoagulated with EDTA and DNA was extracted. The distribution of genetic polymorphism presented by total 15 STR loci D2S1338, D21S11, D7S820, CSF1PO, D3S1358, TH01, D13S317, D16S539, D8S1179, FGA, vWA, TPOX, D18S51, D5S818 and D19S433 from observation and control groups was tested by means of multiplexing PCR-STR, capillary electrophoresis and five-color fluorescent automatic detection. Data analysis used the software SPSS to observe for significant difference between schizophrenia observation group and control group in order to find out the association of schizophrenia with genetic polymorphism presented by 15 STR loci and analyzed the relative risk of significant difference genetic loci. Hardy-Weinberg equilibrium of the observation groups was verified with the goodness of fit test. Twenty people from the schizophrenia observation group, including 10 males and 10 females with averagely aged 46, constitute the observation group for immunological test. Thirty-two blood samples taken from normal people consisting of 16 males and 16 female aged between 40 to 80-year old by Physical Examination Center of Zhong Shan Hospital Affiliated with Dalian University as the control group for immunological test. The test of IgM, IgA and IgG was made respectively for both immunological test groups and the t test was made to analyze for significant difference.Results: The result of test for genetic polymorphism presented by 15 STR loci shown that the STR loci D7S820-11, TH01-7, D19S433-13, D19S433-14.2, D19S433-15, D19S433-15.2 and D18S51-20 from the schizophrenia observation group were obviously different (P<0.05) from those from the control group. The frequency of STR loci D7S820-11, D19S433-14.2 and D19S433-15 was lower than the frequency of those from the control group and the relative risk was less than one. The frequency of STR loci TH01-7, D19S433-13, D19S433-15.2 and D18S51-20 was higher than the frequency of those from the control group and the relative risk was greater than one. The observation value of each genotype of 15 STR loci was well fit with Hardy-Weinberg theoretical expectation (P>0.05), which conformed to Hardy-Weinberg equilibrium. There was significant difference between the two groups in the test of IgM, IgA and IgG.Conclusion: Three short tandem repeats D7S820-11, D19S433-14.2 and D19S433-15 may have a certain relationship with suppression of schizophrenia, and four short tandem repeats TH01-7, D19S433-13, D19 S433-15.2 and D18S51-20 may have a certain relationship with initiation of schizophrenia.
