The Correlation Between Aurora-A Kinase And Breast Carcinoma Treatment Efficacy Of Taxane

Objective: Examine the expression of Aurora-A kinase in mammary gland benign tumor, breast carcinoma and tissues near the tumor, combine with integrated clinical data, analyze its relationship with breast carcinoma treatment efficacy of taxane and relevant prognosis, to provide certain theory basis for breast cancer treatment.Methods: The expression of Aurora-A kinase were tested by immuno- histochemical method S-P in 30 cases of mammary gland benign tumor and 67 cases of breast carcinoma.The expression of Aurora-A kinases together with treatment efficacy of taxane, tumor size, lymph node metastasis, histology graduation and hormone acceptor situation were analyzed statis- tically.Results:1,The expression of Aurora-A kinase in mammary gland benign tumor, breast carcinoma and tissues near the tumor:Aurora-A kinase predominantly localized to the cytoplasm. The expression rate of Aurora-A kinase was 73.13%(49/67) and the overexp- ression rate of it was 55.23%(37/67) in breast carcinoma that was obviously higher than 8.96%(6/67) in tissues near the tumor. There were not expre- ssion of Aurora-A in 30 cases of benign breast tumors2,Relationship between expression level of Aurora-A and treatment efficacy of taxane in breast carcinoma:The treatment effective rate of breast carcinoma was 77.78%(14/18) in negative expression of Aurora-A,rate of nonresponder was 11.11%(2/18); but the treatment effective rate was 34.69%(17/49) in postive expression of Aurora-A,rate of nonresponder was 34.69%(17/49).There were obvious correlations between expression of Aurora-A and treatment efficacy of taxane.Statistical significance can be found between them (P=0.013).3,Correlation between expression level of Aurora-A and the prognosis parameters of breast carcinoma:There were obviously positive correlations between the expression of Aurora-A in breast cancer and the histology graduation, ER and/or PR(P is 0.036,0.007 and 0.014 respectively). That was, the higher the histological grade and ER and PR positive, the higher the level of protein expression. And had not obvious correlation with tumor size, lymph node metastasis, HER-2 expression.Conclusions:1,Aurora-A protein were high expressed in breast carcinoma, low expressed in tissues near the tumor and there was no expression in benign tumor. Examines the expression of Aurora could assist breast cancer treatment before operation.2,There were positive correlations between the expression of Aurora-A in breast cancer and the histology graduation, ER and PR.3,There was manifest correlation between the high expression of Aurora-A and resistance of taxane in breast carcinoma.The testing of Aurora-A can offer theoretical evidence to the medical preparation of breast carcinoma's chemical treatment.
