Porphyrin and their derivative have peculiar physical and chemical character, and play an important role in the photosynthesis and vital process. In recent years, with the development of that macroring drug, the development of porphyrin compounds is very rapid in pharmaceutical research, become one of the advancing topic in porphyrin chemistry, and attract many people to pay attention to them. On the basis of careful study of the most recent documents about the medicine, we have designed and synthesized a series of novel porphyry derivates, and studied their properties in DNA–binding. Provide the experiment basis for the methods of drug molecules and biology macromolecule. The dissertation is included:1. A route to synthesize 5-(p-hydroxy)phenyl-10,15,20- Triphenyl porphyrin(p-HPTPP)follow two kinds method ------Adler and microwave. Results indicate that the method of microwave not only can shorten reaction time, but also can improve productivity.2. Designed the route to synthesize two peptides. By the protection of the amido and the carboxy, combining with DCC and taking off an amine as three fluorine acetic acid, two peptides have been synthesized successfully. Then they have been characterized, assigned and analyzed by ESI-MS, 1H-NMR, IR.3. Designed a series of porphyrin derivate, and synthesized p-NBPTPP, p -CyBPTPP , p-AdBPTPP, p-MPBPTPP, p-SMZBPTPP, p- HOBtBPTPP. All of them have been characterized, assigned and analyzed by ESI-MS, 1H-NMR.4. The interaction betweem porphyrin derivate and DNA have been studied by electronic absorption spectra, viscosity, gel electrophoresis. The results show that these porphyrin derivate have an important effect on DNA and the mechanism of DNA-binding has been discussed.
