Objective: (1)To establish Multi-drug resistant subline HNE-1/ADM in duced from the the human nasophqryngeal carcinoma HNE-1 cell lines .(2) To detect the reversal effect of tetrandrine on multidrug resistance cell line of HNE-1/ADM and investigate its possible mechanisms. (3) To study reversal effect of adriamycin-tetrandrine liposome on the multdrug resistance of the HNE-1/ADM cell line.Methods:(1) A multi-drug resistant cell line HNE-1/ADM human was established by gradually increasing dose of adriamycin and intermittent administration.By means of MT,the growth curves,,multidrug resistance and resistance index of HNE-1/ADM cell line to anticancer agents were detected.By means of microscopy,lectron microscopy and flow cytometry ,the changes of its biological characteristics were determined by light microscopy,electron microscopy,and flow cytometry. (2) MTT reduction assay was used to detect the reversal effect of tetrandrine on HNE-1/ADM cell line;FCM was used to evaluate the effect of tetrandrine on apoptosis of HNE-1/ADM cell line;Immunohistochemisty and Western blot were used to detect the expression of P-gp,Bax and Bcl-2 . (3) MTT reduction assay was used to investigate the reversal effects of adriamycin-tetrandrine liposome and free adriamycin-tetrandrine on the HNE-1/ADM cell line;FCM was used to investigate the apotposis of the HNE-1/ADM cell line by adriamycin-tetrandrine liposome and free adriamycin-tetrandrine ; Immunocytochemistry method and western blot were used to detect the expression of P-gp,Bcl-2 and Bax on the HNE-1/ADM cell line。Results:(1) HNE-1/ADM cell line was established after 4 months,which had a resistance index of 10.913.HNE-1/ADM cells multi-drug resistance to other chemotherapeutic agents.As compared with parental cells,the morphological of HNE-1/ADM were changed;its doubling time was prolonged;and the number of cells in S phase and G0/G1 phase were decreased while in G2/M phase increased.(2) Tetrandrine could reverse drug resistance of HNE-1/ADM cell line to ADM ,and drug resistance index was down regulated from 10.913 to 3.130 . Tetrandrine (1.0μg/ml) couldn't induce the apoptosis of HNE-1/ADM cell ,but could enhance the apoptosis of drug resistance of HNE-1/ADM cell line by ADM . It was observed that the expression of P-gp and Bcl-2 were down regulated ,while,Bax up was up regulated. (3) Adriamycin-tetrandrine liposome was better than free adriamycin-tetrandrine in reversaling the multdrug resistance of the HNE-1/ADM cell line,drug resistance index was down regulated from 10.913 to 1.348; By means of FCM, Adriamycin-tetrandrine liposome was better than free adriamycin-tetrandrine to enhance the apoptosis of drug resistance of HNE-1/ADM cell line,It was observed that the expression of P-gp and Bcl-2 were down regulated , while , Bax up was up regulated by adriamycin-tetrandrine liposome。Conclusion: (1) The typical multidrug resistant phenotype was showed in HNE-1/ADM cell lines. It is a good model for exploring the mechanism of ADM resistant human nasopharyngeal carcinoma cell line .(2) Drug resistance of HNE-1/ADM cell line to ADM can be reversed by tetrandrine, and apoptosis of HNE-1/ADM cell line could be promoted through ADM . It can be postulated that tetrandrine can reverse the drug resistance of HNE-1/ADM cell line by decrease the expression of P-gp and Bcl-2,,increase the expression of Bax.(3) The reversal effects and apoptosis effects of Adriamycin-tetrandrine liposoml in the HNE-1/ADM cell line were stronger them that of Tet. The mechanism showed in this study may be assotionate with the expression of P-gp,Bcl-2,Bax.
