PART 1 EVALUATION OF THE ACTUE CEREBRAL ISCHEMIA-REPERFUSION MEDEL IN RATS USING COMPUTED TOMOGRAPHY PERFUSION IMAGINGObjective: Applicated the CT perfusion imaging (CTPI) techno- logy in Living conditions ,to evaluate its reliability and accuracy in acute cerebral ischemia - reperfusion model .Methods: A stable and reversible acute cerebral ischemia - re perfusion model with unilateral middle cerebral occlusion was established and evaluated by CTPI and TTC staining,HE staining .Results: 1.Both groups CTPI each volume of the various time points between cerebral blood flow (CBF) core areas and cerebral blood volume ( CBV) of low volume infusion areas there were no significant difference (p>0.05), both volumes showed as above there were no significant difference to compare with the TTC staining infarction zone (p>0.05), and there were significant difference to compare with the ischemic volume of mean transit time (MTT) (p<0.05). There were significant differences between MTT ischemic volume and TTC staining infarction zone volume (P<0.05). 2.There were correlation between The CTPI parameters maps and the TTC staining infarct volume maps, and the CBV relevance were the best of them.3. The brain tissue with the matched decrease in CBF and CBV showed the performance-based of nerve cell death in HE staining, the brain tissue with CBF decreased and CBV normal showed the performance-based of nerve cell degeneration.Conclusions: CTPI can evaluate the brain homodynamic of cerebral ischemia-reperfusion animal model in living state as a rapid, accurate, non-invasive method, show the cerebral infarction ischemic region's site, size, extent, and has important clinical value for the ultra-acute cerebral infarction to treatment and review. PART 2 DIAGNOSTIC VALUE OF CT PERFUSION IMAGING IN THE EXPERIMENT OF ULTRA-ACUTE CEREBRAL INFARCTIONObjective: To applicate the technology of CT perfusion imaging (CTPI), observate the dynamic evolution of cerebral blood flow and penumbra at different time points about ultra-acute cerebral infarction, and provide guidance of early diagnosis and treatment for the clinic.Methods: Adult male Wistar rats were divided into ischemia group (n = 24) and sham operation group (n = 6). Each group was executed of CT perfusion scan and HE staining respectively and in series at 1, 2h, and 3,4,5,6 h after surgery.Results:â’ˆCTPI and HE staining showed negative of the sham group.â’‰CTPI performance of the ischemic group:â‘ At the central area of infarction ,the CBF and CBV was lower than the opposite, MTT was prolonged, there were a significant difference (p <0.05);The CBF significantly lower than the opposite at Penumbra, there was a significant difference (p <0.05).â‘¡At the central area of ischemic infarction, the rCBF, rCBV and MTT changed significantly at 1h, 2h compared with the 3h, 4h, 5h, 6h, there was a significant difference (p <0.05).â‘¢There was a significant difference of rCBF,rCBV,MTT, the central area of infarction compared with the penumbra, the area around the normal comparison, (p <0.05);And there was a significant difference of rCBF about Penumbra compared with the area around the normal (p <0.05).â‘£The infarction area of the central expanded significantly over time, and there was a significant difference 1h, 2h compared with 3h, 4h, 5h, 6h (p <0.05).â’ŠHE staining performance of the ischemic group: At 1h, 2h, 3h point , blood vessels gap widened , the number of neurons reduced, the nerve cells edema mild; At4h, 5h, 6h point, widened significantly of vascular space and neurons gap, condensated of the nucleus, and there were a great quantity of vacuoles around nerve cells and blood vessels .Conclusion: The check of CTPI was non-invasive, simple and quick, and it could show the scope and site of cerebral infarction and penumbra at ultra-acute period, the hemodynamic changes with quantitative analysis, and reduced the treatment time window significantly, provided a great role for clinical treatment. In particular, there was a great significance about the penumbra imaging technology for the research of therapeutic time window and drug. PART 3 IMAGING APPLICATION FOR THE EXPE-RIMENT OF MARROW STROMA CELLS IN THE TREATMENT OF ISCHEMIC STROKEObjective: To investigate the value of imaging application for the experiment of Mesenchymal stem cells transplantation in the treatment of ischemic stroke.Methods: Adult male Wistar rats were divided into sham operation group , ischemic reperfusion (MCAO )group, PBS treated group and MSCs treated group, and at 1d,3d,7d, 14d, 28d,each was executed examination of CTPI,MRI,Neurological score,TTC staining,HE staining and microvessel density analysis.Results:â’ˆThere was no infarction and hemodynamic was normal on each point in Sham group .2. MCAO group, PBS group, MSCs group:â‘ There was no significant difference about the volume of infarction about CBF,CBV maps of CTPI,T2WI images compared with TTC staining; 1d,3d compared with 7d,14d,28d, there was a significant difference,P<0.05;â‘¡MSCs group compared with MCAO group, PBS group, rCBF increased on the point of 7d,14d,28d , there was a significant difference ( P<0.05) , and Neurological score lowed,microvessel density raised ,TTC staining showed cell degeneration and necrosis reduced .â’Šlong-T1, short-T2 signal could be seen around the infarction at 3d,7d,the Low signal diffused to the direction of the infarction and the intensity reduced in MSCs group.Conclusion: The use of CTPI and MRI could observe the effects of MSCs in the treatment of ischemic stroke about morphology and homodynamics, also could track magnetic labeled cells, survival, migration and differentiation in living with MRI, and then provided a broad platform for stem cell transplantation research.
