Objective: To investigate the expression of aquaporin5(AQP5) and extracellular signal-regulated kinase protein(ERK) and their correlation in colorectal carcinoma.Methods:The differential protein expression of AQP5 and ERK between 60 samples of colorectal carcinoma and 20 samples of its corresponding normal colorectal mucosa was analyzed by microtissue array and immunohistochemistry. Moreover, The AQP5 and ERK protein expression were further analyzed by Western blotting hybridization in 15 colorectal carcinoma tissue versus the counterparts.Results:â‘ The expression of AQP5 and ERK protein in colorectal carcinoma by immunohistochemistry was higher than in the corresponding normal colorectal mucosa. Both expression of AQP5 and ERK protein were related with the depth of tumor invasion, lymph node metastasis and TNM stage(P<0.05).â‘¡The results of Western blotting suggested that of 15 paired samples, AQP5 and ERK protein was up-regulated in colorectal carcinoma compared with their normal colorctal mucosa in all the cases(P<0.05).Further, semiquantitive protein expression of AQP5 and ERK were also related with the depth of tumor invasion, lymph node metastasis and TNM stage(P<0.05).Moreover, the expression of AQP5 protein was positively related with the expression of ERK proein in colorectal carcinoma(P<0.05).Conclusions: These findings indicate that expression level of AQP5 and ERK protein is closely related to the biological behavior of colorectal cancer. The expression level of AQP5 protein is closely associated with the overexpression of ERK protein. AQP5 protein may be able to promote Ras/Raf/MEK/ERK signaling pathways, resulting in invasion and metastasis.
