Obsjective:To explore the expression of receptor for advanced glycation end products (RAGE) and type IV collagen(Col4) in ovary tissue of women with and without polycystic ovary syndrome(PCOS).To analyze the relationships of RAGE and Col4 to body mass index(BMI), waist to hip ratio(WHR), fasting plasma glucose (FPG), fasting insulin(FINS), insulin resistance index and the levels of serum sexual hormones. To explore the fuction of RAGE and Col4 in PCOS.Methods:Samples from PCOS(n=40)and controls(n=20) were collected.1. Gene expression of RAGE and Col4were detected by reverse transcriptase polymerase chain reaction(RT-PCR) in the ovary tissues of the two group.2. Serum follicle stimulating hormone(FSH),luteinizing hormone(LH), estradiol(E2), testosterone(T), prolaction(PRL) and fasting insulin level(FINS) were measured by chemiluminescence assay.Fasting plasma glucose(FPG) were measured by glucose oxidase method.3. Insulin resistance index was calculated using homeostasis model assessment insulin resistance(HOMA-IR).4. The relationships of RAGE and Col4 to body mass index(BMI),waist to hip ratio(WHR),fasting plasma glucose(FPG),fasting insulin(FINS),insulin resistance index and the levels of serum sexual hormones were analyzed by correlation and regression.Results:1. There was significant up-regulation of RAGEmRNA in ovary tissue in women with PCOS when compared with normal controls[ ( 2.29±0.24 ) vs(1.89±0.23), P=0.001].2. Greater amounts of Col4mRNA are present in the ovary tissues of women with PCOS when compared with normal controls[ ( 1.15±0.23 ) vs(0.88±0.19), P=0.001]. 3. FINS,HOMA-IR,LH,LH/FSH and T were also significantly greater in women with PCOS(P<0.05).4. RAGEmRNA expression in ovary tissue was positively correlated with BMI (r=0.33, P=0.01), WHR(r=0.309, P=0.016), FINS(r=0.342, P=0.008), HOMA-IR (r=0.556,P=0.001),T(r=0.566,P=0.001) and Col4mRNA expression in ovary tissue(r=0.344,P=0.007). Col4mRNA expression was positively correlated with FINS(r=0.305,P=0.009),HOMA-IR(r=0.267,P=0.02) , T(r=0.398,P=0.001) and RAGEmRNA expression in ovary tissue (r=0.344, P=0.007).5. Multiple regression analysis showed that T(P=0.003)and HOMA-IR(P=0.005) were relatiaed factors in influencing RAGEmRNA expression. T(P=0.002) was related factor in influencing Col4mRNA expression.Conclusion:1. The expression of RAGE and Col4 in the ovary tissue are up-regulation in women with PCOS.2. The expression of RAGE is related to insulin resistance and hyperandrogenemia.3. The expression of Col4 is related to hyperandrogenemia.
