ObjectiveTo detect the expressions of advanced glycation end products (AGEs)and its receptor(RAGE)in the retina of diabetic rats treated withpyridoxamine, to study the protective effect of Pyridoxamine on theretina of Diabetic Rat.Methods6weeks old male SD rats (clean grade) were randomly divided into fourgroups: normal control group (NC group), diabetes group (DM group),diabetes treated with pyridoxamine group (PM group) and diabetes treatedwith aminoguanidine group (AG group). Type1Diabetic rats were inducedby an intraperitoneal injection of streptozotocin (STZ). NC group andDM group were treated with distilled water intragastric administration(2ml/d),PM group was treated with pyridoxamine intragastricadministration(200mg·kg-1·d-1),and AG group was treated withaminoguanidine intragastric administration(100mg·kg-1·d-1).4and12weeks later,the rats were killed and the eyeballs were removed.Theexpressions of AGEs in serum and retina were measured by enzyme-linkedimmunosorbent assay (ELISA). The retina distributions of RAGE weredetected by immunofluorescence staining and the mRNA expressions of RAGEwere quantified by Real-time polymerase chain reaction (RT-PCR)respectively. Results4and12weeks later, the content of AGEs in serum and retinal of PMgroup was significantly lower than DM group and AG group(P<0.01); ineach group the content of AGEs at12weeks was increased significantlymore than4weeks(P<0.01).The expression of RAGE was increased anddetected from retinal pigment epithelium to ganglion cell layer byimmunofluorescence staining of diabetic rats. After treatment withpyridoxamine,The RAGE expression decreased significantly and its ratiowas obviously lower than that of the DM group and AG group.ConclusionsPyridoxamine can significantly down-regulate the expressions of AGEsand RAGE in serum and retina of diabetic rats.It may protect the retinafrom the impairment of AGEs and AGEs-RAGE system....
