| Objective To study the inhibition effect of Breast-cancer Metastasis Suppressor 1 on primary tumor growth and metastasis of human gastric cancer cells in nude mice.Methods Cultured human gastric cancer cell line SGC-7901,the cells were divided into 3 groups:transfected group with pcDNA3.1(-)B/myc-BRMS1;negative control group with pcDNA3.1/myc-his(-)B;blank control group without any transfection;The stability expression stain was screened by the condition media containing 400 mg/L G418 and cloned through the limited dilution method;The mRNA of BRMS1 were evaluated by means of RT-PCR;Histologically intact tissue of 3 groups of cells maintained by passage in the subcuitis of nude mouse was transplanted orthotopically into the stomach wall of nude mice to establish the nude mouse model of human gastric carcinoma,then their primary tumor growth and metastasis were observed.Results The mRNA of BRMS1 were expressed in transfected group cell s by means of RT-PCR,and there were not detected the product of BRMS1 gene primers in negative control group and blank control group;There were no significant differences among transfected group 12.02±0.70mm,n egative control group 12.71±0.63mm and blank control group 12.89±0.71mm(P＞0.05) in the size of orthotopic tumor in nude mice; The numbers of metastasis tumor in transfected group,negative control group and blank control group were 4.38±0.60,7.75±0.59, 7.63±0.65 respectively;with significant differences among 3 group s(P＜0.05).Conclusion BRMS1 gene didn't affect gastric tumor growth,but suppress metastasis in nude mice.
